Rapid Separation of Indole Glucosinolates in Roots of Chinese Cabbage (Brassica rapaSubsp. Pekinensis) by High-Performance Liquid Chromatography with Diode Array Detection

Abstract: Glucosinolates are a class of sulphur-containing plant compounds with diverse biological properties. They have been found exclusively in the Brassicaceae family plants and studied exhaustively in biosynthetic and application perspectives. The aim of this current study is to present a simple and updated method to quantify indole glucosinolate content in hairy root cultures of Chinese cabbage by HPLC-DAD-UV/Vis. Method validation controls were performed and recovery experiments were assayed. The data was statica… Show more

“…Glucoraphanin, with the basic structure of glucosinolates, consists of sulfur-bonded β- d -glucopyranose, sulfonated oxime, and side chain (R group) derived from different amino acids. When hydrolysis removes β- d -glucopyranose, the remaining structure spontaneously undergoes Lossen rearrangement to remove sulfate ions and form a metabolite . Therefore, the principles of bacterial metabolism of different glucosinolates are similar.…”

“…In the leaves (L) and the roots (R) collected, we detected the GLSs total amount and their profile. The extraction method of the GLSs was based on the International Standard Method ISO 9167-1, 1992 [37], which the European Commission has formally adopted (European Commission, 1990), with several modifications [38]. An amount of 200 mg of the freeze-dried samples was boiled in 5 mL of methanol 70% for 10 min at 70 • C to inactivate myrosinase, thereby preventing the enzymatic hydrolysis of the GLSs.…”

“…However, this number was later reconsidered due to insufficient documentation (Blažević et al, 2020). Aires and Carvalho (2017) suggested a simple method to quantify indole glucosinolate by HPLC-DAD-UV/Vis. A comprehensive critical review of the diversity of plant glucosinolates is thoroughly discussed by Blažević et al (2020) where glucosinolate structures were fully or partially Core chemical structure of glucosinolates (top) and different side chains as the R group of the most representative glucosinolates found in nature.…”
