Rapid, Low-Technology MIC Determination with Clinical Mycobacterium tuberculosis Isolates by Using the Microplate Alamar Blue Assay

Abstract: A colorimetric, microplate-based Alamar Blue assay (MABA) method was used to determine the MICs of isoniazid (INH), rifampin, streptomycin (SM), and ethambutol (EMB) for 34 PeruvianMycobacterium tuberculosis isolates (including both pansensitive and multidrug-resistant strains) and the H37Rv strain by using bacterial suspensions prepared directly from solid media. Results for all isolates were available within 8 days. Discordant results were observed on initial tests for 3 of 16 INH-susceptible isolates, 5 of … Show more

“…Plates were not counted if microscopic growth was so heavy that individual colonies could not be identified. The microplate Alamar blue assay was used to determine the efficacy of STD and DPPC LE-doxy in a cell-free system as a comparison (Franzblau et al, 1998).…”

“…The compounds were tested at concentrations of 1, 5, 10, 20 and 40 lg/mL. The minimal inhibitory concentrations (MIC 99 ) of compounds 1-20 were determined using the proportion method in the BACTEC MGIT 960 system (9) in the TB Exist mode (24). Compounds 2, 3, 6, 7, 10, 15-17, and 19 all had a negative effect on the growth of mycobacterial cells, with 99% inhibition observed at concentrations between 5 and 40 lg/mL.…”

“…All the newly synthesized peptides 1-11 were evaluated in vitro antitubercular screenings against MTBH37Ra using the microplate Alamar blue assay (MABA) method. The activity is expressed as % inhibition at various concentrations used for the assay (Franzblau et al, 1998;O'Brien, Wilson, Orton, & Pognan, 2000). Compounds were suspended in Milli-Q water (Stock con.…”
