Rapid identification of important periodontal microorganisms by cultivation

Abstract: Two recent developments have made possible rapid isolation and identification of important periodontal species. Selective media have been designed which demonstrate high recovery efficacy of black‐pigmented Bacteroides spp., Actinobacillus actinomycetemcomitans, Eikenella corrodens, Fusobacterium spp., Capnocytophaga spp. and other periodontal bacteria. Presumptive tests for identification of periodontal isolates that are rapid and easy have also been described. Available microbiological methods permit even sm… Show more

“…The Trypticicase Soy Serum Bacitracin Vancomycin agar (TSBV) medium was incubated in 10% CO 2 at 37ºC for 4 days. Presumptive identification was performed according to the methods described (14), and using a commercial identification micromethod system (RapID ANA II, Remel, Norcross, GA, USA) for A. actinomycetemcomitans and P. gingivalis. Total viable counts (TVC) were defined as the total number of colonyforming units obtained on non-selective media plates.…”

“…Generally, isolation of microorganisms was carried out by methods previously reported by D' Ercole et al (2008). The samples were analyzed using microbial culture techniques for the presence of periodontopathic bacteria according to Slots (1986). All samples were processed in ≤24 hours at room temperature and immediately incubated in CO2 and anaerobic culture systems.…”

“…The trypticase-soy with serum, bacitracin, and vancomycin medium was incubated in 10% CO2 in air at 37ºC for 4 days. Presumptive identification was performed according to the methods described (Slots;Slots & Reynolds, 1982) and using a commercial identification micromethod system (RapID ANA, Remel, Norcross, GA). Total viable counts (TVC) were defined as the total number of colony-forming units obtained on non-selective media plates.…”

“…Las cajas de agar Brucella se incubaron en anaerobiosis por siete días para la recuperación de los demás periodontopatógenos. La identificación se hizo teniendo en cuenta características morfológicas de las colonias y pruebas bioquímicas adicionales (14). Para identificar las levaduras se subcultivó la colonia presuntiva en agar TSBV al agar sangre evaluándose el aspecto de la colonia y la morfología microscópica correspondiente a blastoconidias en la preparación con azul de lactofenol (15).…”
