Rapid glucocorticoid inhibition of vasoactive intestinal peptide-induced cyclic AMP accumulation and prolactin release in rat pituitary cells in culture.

Abstract: Vasoactive intestinal peptide (VIP) stimulates both adenosine 3',5'-cyclic monophosphate (cAMP) accumulation and prolactin release in normal rat pituitary cells in culture. cAMP accumulation is significant (P < 0.01) at VIP concentrations as low as 1 nM and reaches a maximum with 0.1 ,IM. Addition of dexamethasone as early as 15 min before VIP inhibits VIP stimulation of both cAMP production and PRL secretion. The rapid inhibition is dose-dependent: it appears at doses as low as 0.01 pM and is complete at 1 pM… Show more

“…As reported in [6,8], under similar experimental conditions, VIP induced a dose-dependent increase in cAMP accumulation and PRL release with a maximum increase occurring at 10-7 M. Increasing [SRIF] from 10-10-10 6 M inhibited in a dose-dependent manner the stimulation of both cAMP accumulation and PRL secretion induced by 10 7 M VIP ( fig.l). Half-maximal inhibition for both cAMP accumulation and PRL secretion was obtained with -1-2 × 10-1° M SRIF.…”

“…In [7,8] VIP receptors in the pituitary were coupled to cAMP, and the increase of cAMP accumulation was related to stimulation of PRL secretion [7,8]. However, conflicting data have been reported regarding the involvement of cAMP in the mechanism of action of SRIF [13].…”
