Rapid diagnosis of tuberculosis directly from clinical specimens using a gene chip

Abstract: The aim of this study was to explore a gene chip capable of detecting the presence of Mycobacterium tuberculosis isolates directly in clinical sputum specimens and to compare it with current molecular detection techniques. At first, we selected 13 M. tuberculosis-specific target genes to construct a gene chip for rapid diagnosis. Using the membrane array method, we diagnosed M. tuberculosis by gene chip directly from 246 sputum specimens from patients suspected of having tuberculosis. Among 80 M. tuberculosis … Show more

“…[10][11][12] A variety of microfluidic devices have been fabricated based on this microfabrication process, such as micropumps, 13 microvalves, 14 microseparators, 15 microconcentrators, 16 and micromixers. 17 Furthermore, these PDMS-based microfluidic devices can even be integrated to perform a specific assay for various applications such as molecular diagnosis, 18 protein analysis, 19 and cell studies. 20 However, there are still potential limitations for PDMS-based microfluidic systems.…”

“…The advent of molecular biology techniques that led to significant advances in DNA amplification and hybridization techniques has significantly aided the rectification of current flaws in the diagnosis of cutaneous mycobacterial infections ( Jagielski et al., 2016 ). DNA sequencing, PCR-restriction fragment length polymorphism (RFLP) assays ( Wong et al., 2003 ), gene chip assays ( Chang et al., 2010 ), and commercial kits, such as GenoType and INNO-LiPA, are established nucleic acid-based assays for identification of mycobacterial infections ( Zhu et al., 2010 ). DNA sequencing, especially metagenomic next-generation sequencing (mNGS) ( Kong et al., 2022 ), has greater efficiency, sensitivity, and specificity in detection of pathogenic bacteria, and can even detect rare pathogens.…”

“…Previously, the instrumentation and microarray components had been independently evaluated for detection of other infectious entities, such as staphylococcal isolates [21]. Recently, the technology of microarray had been implemented in identification of MTB complex [22], [23], which gave rapid and accurate detection of MTB complex. Meanwhile, microarray had been used in drug resistance TB finding in near years by [14], [24].…”
