Rapid diagnosis of rabies in humans and animals by a dot blot enzyme immunoassay

Abstract: The dot blot enzyme immunoassay (DIA) test described here is a sensitive, specific and rapid test for the post-mortem diagnosis of rabies in animals and humans. The utility of this test for the ante-mortem diagnosis of rabies needs to be further evaluated.

“…Positive reactions were easily visualized as red dots after enzyme degradation of the substrate. Dot-ELISA test for rabies detection was found to be specific, faster and reliable with almost 92% sensitive which is similar to the earlier reports [18,24]. Although, Dot ELISA test did not produce non-specific false-positive results [19], development of colour spot depends on the concentration of virus in antigen used in the test.…”

“…The bacterial suspension was blotted directly onto nitrocellulose membranes using a 96-multiwell microfiltration blotter (BioRad Laboratories, Hercules, Calif., U.S.A.) (1,6,12,14). After blocking with 10% skimmilk in PBS for 30 min, the membranes were exposed to 1 mg/ml of PlyGB-GST for 60 min and then 200 ng/ml horseradish peroxidase (HRP)-conjugated mouse anti-GST mAb (GST (B-14) HRP; Santa Cruz, Calif., U.S.A.) for 60 min.…”
