Rapid Detection of Viral Envelope Lipids Using Lithium Adducts and AP-MALDI High-Resolution Mass Spectrometry

Abstract: There is an unmet need to develop analytical strategies that not only characterize the lipid composition of the viral envelope but also do so on a time scale that would allow for high-throughput analysis. With that in mind, we report the use of atmospheric pressure (AP) matrix-assisted laser desorption/ionization (MALDI) high-resolution mass spectrometry (HRMS) combined with lithium adduct consolidation to profile total lipid extracts rapidly and confidently from enveloped viruses. The use of AP-MALDI reduced … Show more

“…The inlet capillary was set to 400 °C. 25 Each dried-droplet spot was scanned with a 50 μm wide laser spot. Mass spectra were acquired in a positive-ion mode with mass resolving power up to 180 000 at m / z 200 and a mass range of m / z 150–1500.…”

“…Eight subclasses, including 132 kinds of lipids were identified. 73 The construction of the AP MALDI selected reaction monitoring (SRM) method by Vishal Mahale's group in 2022 provided a rapid and quantitative platform for analysis of aflatoxin M1 in milk. The calibration curve displayed excellent linearity ( R 2 = 0.99) with good recoveries for quality control samples (97–106%) and this method's property was validated by ultrahigh-performance liquid chromatography with fluorescence (UHPLC-FLD).…”

“…17,18 This is due to both the relative ease of ionization of several common lipid classes as well as the central role of lipid metabolism in countless biological processes and diseases. 2,19,20 MALDI analysis is conventionally performed under vacuum, however, atmospheric pressure (AP) arrangements are also used, 21,22 having advantages including ease of sample exchange and a broader tolerance for analysis of volatile analytes and vacuum incompatible samples. In the context of lipid analysis, AP-MALDI approaches can also be advantageous for labile lipid species, such as gangliosides, that show reduced in-source fragmentation when ionized at AP.…”
