“…Rainbow trout (ca 12 g) were obtained from a freshwater commercial farm located in a n area where piscirickettsiosis has never been reported (Metropolitan Region of Chile). To confirm the absence of Piscirickettsia salrnonis, kidney smears were tested by an indirect fluorescence antibody test (IFAT) according to the method of Lannan et al (1991), as modified by Larenas et al (1996a). A polyclonal P. salmonis antiserum, kindly provided by Mrs C. N. Lannan, from Oregon State University (USA) was used.…”

“…The fluorescent antibody test can be used to confirm the presence of P. salmonis in smears and impressions directly from infected fish tissues, in suspect tissue cultures, and in paraffin‐embedded tissue sections (Lannan, Ewing & Fryer 1991; Larenas, Astorga, Contreras, Garcés, Fryer & Smith 1996a; Jamett et al. 2001).…”

“…Appropriately fixed preparations are incubated with rabbit anti‐ P. salmonis serum or mouse monoclonal antibodies and then with fluorescein or other dye‐conjugated antibodies directed to rabbit IgG. A similar test can be applied to deparaffinized tissue sections from infected fish with better results obtained if the sections are subjected to microwave irradiation to reveal P. salmonis antigens (Larenas et al. 1996a).…”
