Rapid and generic identification of influenza A and other respiratory viruses with mass spectrometry

Abstract: The rapid identification of existing and emerging respiratory viruses is crucial in combating outbreaks and epidemics. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) is a rapid and reliable identification method in bacterial diagnostics, but has not been used in virological diagnostics. Mass spectrometry systems have been investigated for the identification of respiratory viruses. However, sample preparation methods were laborious and time-consuming. In this study, … Show more

“…At present, mass spectrometry has had limited application in clinical virology6232425. In this light, the main purpose of this study was the development of a new tool based on MALDI-TOF MS as a model to be applied to the identification of cultivable respiratory viruses after a cell culture step.…”

“…Despite many literature data about MALDI-TOF MS application in routine clinical microbiology, and in experimental approaches related to bacteria, parasites, yeasts and moulds616171819202122, very few reports are available about its application in virus research and laboratory diagnosis of viral infections6232425. In most of the studies, specific viral genome sequences are previously amplified by PCR and then the amplicons are analysed by MALDI-TOF MS15.…”

“…From these peptide sequences, not only proteins of interest can be identified, also the micro organism itself can be specified to, at least, species level [16,17]. Recent developments in proteomic analysis using LC-MS/MS reduced the time to result, making it possible to analyze a sample within 6 h [18].…”

“…The potential for clinical applications of MS for studying host cell interaction and analyzing biomarkers of the virus itself or linked to the host response has been reviewed elsewhere . The detection of viral proteins can be performed directly in various fluids or cells from infected patients . However, viral identification suffers from their highly evolutive nature.…”
