Rapid and Efficient Ambient Temperature X-ray Crystal Structure Determination at Turkish Light Source

Abstract: High-resolution biomacromolecular structure determination is essential to better understand protein function and dynamics. Serial crystallography is an emerging structural biology technique which has fundamental limitations due to either sample volume requirements or immediate access to the X-ray beamtime. Obtaining a high volume of well-diffracting, sufficient-size crystals while mitigating radiation damage remains a critical bottleneck of serial crystallography. As an alternative, we introduce the plate-read… Show more

“…N16961( Vc ) which possesses the catalytic cofactor FAD, an antenna chromophore DMRL, and an additional cofactor [4Fe-4S] cluster ( 15 ). We determined the crystal structure of Vc (6-4) PL at 2.5 Å resolution at cryogenic temperature at the Turkish Light Source known as “ Turkish DeLight ” ( 24 , 25 ). The structure contains all the cofactors with well-defined electron densities and contains a total of 24 α helixes and five β sheets ( Fig.…”

“…The crystal belongs to the P1 triclinic space group with a=45.885, b=51.7048, c=54.3003, α=91.04, β=112.15, γ=108.43. The dimerized structure of TRAF6 N-terminal region was determined to 2.6 Å resolution at cryogenic temperature at the Turkish Light Source “ Turkish DeLight ” [40] (Figure 1). The determined structure was deposited to PDB database with the ID: 8HZ2.…”
