Ranunculus latent virus: a strain of artichoke latent virus or a new macluravirus infecting artichoke?

Abstract: An elongated virus was isolated from artichoke crops in Liguria, and a 700-bp fragment was amplified by RT-PCR using oligonucleotides to detect members of the family Potyviridae. Comparison of fragment sequences showed 98% identity at the nucleotide level with the ranunculus isolate of the macluravirus Ranunculus latent virus (RaLV). RaLV was then detected by DAS-ELISA in symptomatic and asymptomatic artichoke plants from Liguria, Sardinia and Latium. The sequence of a 5.5-kb region was assembled from a cDNA l… Show more

“…Subsequently, other members infecting economic plants were identified and currently, six species are officially recognised (http://www.ictvonline.org/virusTaxonomy.asp?version=2009). In addition to LCCV reported in this study, two more new species Yam chorotic necrotic mosaic virus (Wang et al , ) and Artichoke latent virus (Ciuffo et al , ) were reported during last 2 years. There is a rising importance of the macluraviruses as a taxonomic group and it is necessary to revise the genus Macluravirus .…”

“…The denatured CP of the purified virus showed two bands of about 40 and 35 kDa, of which 40 kDa is the estimated molecular mass of the capsid protein as the lower band is expected due to proteolytic degradation as seen in CdMV (Gonsalves et al , ; Jacob & Usha, ). Contrastingly, in RaLV no degradation but multimerisation of CP was obtained above the 40‐kDa band (Ciuffo et al , ). The deduced aa sequence, however, showed a lower molecular mass of 33.7 kDa.…”

“…Occurrence of virus-like symptoms prompted surveys (107,109,130) along the Italian Riviera, where Ranunculus is cultivated for export. Previously reported viruses and three new potyviruses were identified: ranunculus leaf distortion virus, ranunculus mild mosaic virus, and ranunculus mosaic virus; a new macluravirus, artichoke latent virus (20), was also detected. Ranunculus white mottle virus (RWMV), a new ophiovirus detected in about 30% of ranunculus plants, was also found in anemone (132).…”

“…Ranunculus hybrids, subsequently referred to as ranunculus, are susceptible to infection by several viruses, in most cases found in mixed infection, all associated with economically important diseases (Vaira et al ., ; Garibaldi et al ., ; Turina et al ., ; Ciuffo et al ., ; Restuccia et al ., ). Common viral symptoms on ranunculus are mottling, chlorosis, mosaic, necrosis and deformation on leaves, vein yellowing, stunting of the plant and deformation and colour breaking on flowers.…”
