RANBP1 (RAN Binding Protein 1): The Missing Genetic Piece in Cancer Pathophysiology and Other Complex Diseases

Abstract: RANBP1 encoded by RANBP1 or HTF9A (Hpall Tiny Fragments Locus 9A), plays regulatory functions of the RAN-network, belonging to the RAS superfamily of small GTPases. Through this function, RANBP1 regulates the RANGAP1 activity and, thus, the fluctuations between GTP-RAN and GDP-RAN. In the light of this, RANBP1 take actions in maintaining the nucleus–cytoplasmic gradient, thus making nuclear import–export functional. RANBP1 has been implicated in the inter-nuclear transport of proteins, nucleic acids and microR… Show more

“…RanBP1 is known to act as an adjuvant to Ran and to be involved in mitosis and in the cell cycle [ 30 ]. RanBP1 shows the highest activity in the early and late stages of the cell cycle, in order to help Ran function [ 31 ]. Additionally, RanBP1 plays an essential role in the directional migration of neural crest cells during development [ 32 ].…”

“…IL-18, regulated by RanBP1, was discovered 30 years ago and causes a variety of biological effects [ 31 ]. We found that IL-18 re-regulates RanBP1 to form an autocrine loop.…”

“…We hypothesise asthma might in uence ROS homeostasis and innate immunity of CRSwNP, which needs to be explored further. RanBP1 is a certain cell cycle regulatory protein belonging to the regulator of the Ran-GTPase, and plays regulatory functions in the RAN network 24 . Transforming growth factor-β (TGF-β), which is particularly important in promoting remodeling in airway diseases like asthma and CRSwNP 25 .…”

“…Compared to HIV-1, HIV-2 managed to downregulate the expression of RANBP1, which deserves significant attention. This protein was found to contain a nuclear export signal (NES) domain that is similar in structure and function to that found in HIV Rev; therefore, it is hypothesized that it shares a step in the post-transcriptional pathway with Rev [ 90 ]. Its downregulation by HIV-2 raises questions about whether it could be implicated in the suppression of the transport of proviral transcripts.…”
