Radioprotective Agents and Enhancers Factors. Preventive and Therapeutic Strategies for Oxidative Induced Radiotherapy Damages in Hematological Malignancies

Abstract: Radiation therapy plays a critical role in the management of a wide range of hematologic malignancies. It is well known that the post-irradiation damages both in the bone marrow and in other organs are the main causes of post-irradiation morbidity and mortality. Tumor control without producing extensive damage to the surrounding normal cells, through the use of radioprotectors, is of special clinical relevance in radiotherapy. An increasing amount of data is helping to clarify the role of oxidative stress in t… Show more

“…These activations are responsible for radioresistance in cancer ( 85 ). The inhibition of oxidative stress appears to be the main mechanism, established by the intracellular antioxidant system, responsible for tumor radioresistance ( 121 ). As presented in Table II , increasing evidence has demonstrated that antioxidant system inhibitors promote radiation sensitization.…”

“…IR countermeasures protect organisms against the harmful effects of IR and reduce tissue damage ( 11 ). Based on the time of administration relative to IR exposure, countermeasures against radiation injury are classified as radioprotectors, radiation mitigators or therapeutics ( 12 15 ).…”

“…IR can directly hit DNA or indirectly by increasing the generation of free radicals, i.e., oxidative stress ( Leroi et al, 2016 ; De Ruysscher et al, 2019 ; Allegra et al, 2020 ). The cytotoxic effects of IR on normal cells are essentially due to the radiolysis of cellular water, leading to instant ionization of water molecules, producing free radicals including reactive oxygen species (ROS) and nitrogen species (RNS), such as hydrogen peroxide (H 2 O 2 ), superoxide anion (O 2 •- ), hydroxyl radical (OH • ), and nitric oxide (NO).…”
