Radioiodination of balsalazide, bioevaluation, and characterization as a highly selective radiotracer for imaging of ulcerative colitis in mice

Abstract: This work focuses on tracking ulcerative colitis in mice. High labeling yield and radiochemical purity were achieved for the formation of a [125/131I]balsalazide radiotracer at optimum conditions of oxidizing agent content (chloramines‐T [Ch‐T], 75 μg), substrate amount (100 μg), pH of reaction mixture (6), reaction time (30 min), and temperature (37°C), using radioactive iodine‐125 (200–450 MBq). The radiolabeled compound, [125/131I]balsalazide, was stable in serum and saline solution during 24 h. Balsalazide… Show more

“…The total yield of the radiotracer consists of free [ 125 I] -iodide and [ 125 I] -ZEN it is calculated from the percentage (%), on TLC at RTs (retention times) 0.0 and 0.9–1.0 respectively. HPLC analysis revealed the following, the purity for [ 125 I]-ZEN reached 99 % [ [101] , [102] , [103] , [104] , [105] , [106] , [107] , [108] , [109] , [110] , [111] , [112] , [113] , [114] , [115] , [116] , [117] , [118] , [119] , [120] ]. The Retention times values for both the free iodide and the labeled molecule [ 125 I]-ZEN existed at 4.5 and 15.8 min, respectively, with regard to the retention time for ZEN that was 14.7 min ( Fig.…”

“…A standard deviation (SD) for the mean value of the percentage of the supplied dose per gram was determined. It was calculated that muscle made up 40% of a person's body weight, whereas bone made up 10%, and blood made up 7% [71][72][73][74][75][76][77][78][79][80]. Experiments were conducted while corrections were used for background radiation and emission decays.…”

“…2B). By studying the effect of pH in the range 2-15, it was discovered that pH 8 is the optimum value for achieving the highest labelling yield of [ 125 I] iodoolsalazine radiotracer [84]. Furthermore, the effect of the reaction time and oxidizing agent (Ch-T) amount was investigated to obtain the highest labelling yield (98.5%) at 30 minutes and 100 μg, respectively (Fig.…”
