A B S T R A C T The secretion of androgens and estrogens by normal and abnormal testes was compared by determining the concentrations of dehydroepiandrosterone (DHEA), androstenedione (A4A), testosterone (T), estrone (El), and 17f-estradiol (E2) in peripheral and spermatic venous plasma samples from 14 normal men and 5 men with unilateral testicular atrophy. Four normal men and one patient with unilateral atrophy of the testis were given human chorionic gonadotropin (HCG) before surgery. Plasma estrogens were determined by radioimmunoassay; plasma androgens were measured by the double-isotope dilution derivative technique. Peripheral concentrations of these steroids before and after HCG were similar in both the normal men and the patients with unilateral testicular atrophy. In normal men, the mean +SE spermatic venous concentrations were DHEA, 73.1±+11.7 ng/ml; A4A, 30.7±7.9 ng/ml; T, 751±114 ng/ml; Ei, 306±55 pg/ml; and Ea, 1298±216 pg/ml. Three of four subjects with unilateral testicular atrophy had greatly diminished spermatic venous levels of androgens and estrogens. HCG treatment increased In the single subject with an atrophic testis who received HCG, the spermatic venous concentrations of androgens and estrogens were much less than in normal men similarly treated. We conclude that: (a) El is secreted by the human testis, but testicular secretion of El accounts for less than 5% of El production in normal men; (b) HCG stimulation produces increases in spermatic venous estrogens equal to or greater than the changes in androgens, including testosterone; and (c) strikingly decreased secretion of androgen and estrogen by unilateral atrophic human testes cannot be appreciated by analyses of peripheral steroid concentrations.

SUMMARYThe role of androgens and estrogens has been investigated in postmenopausal breast cancer patients. No differences were found in the urinary excretion of estrone and of estriol between 41 primary mammary cancer patients and 48 normal postmenopausal women, representative of the normal population. A significantly lower excretion of androgen metabolites (ll-DOKS) was found in the patients. Because ll-DOKS in postmenopausal women arise mainly from three secretory products and estrogens are mainly derived from peripheral conversion of androstenedione to estrone, production rates of DHEA, DHEAS and androstenedione and the conversion of androstenedione to estrone were estimated. No significant difference was found between the two groups for the blood production rate of androstenedione, neither for its conversion to estrone. The urinary production rate of DHEAS was definitely lower in the selected breast cancer patients compared to normal controls. The DHEA production rate was also lower but statistical significance was not achieved.On account of these results the hypothesis was tested that DHEAS, DHEA or one of their metabolites might interfere with the binding of estradiol to its specific receptor, an essential step in its mechanism of action. From the results of an in vitro incubation study of receptors from human myometrial and mammary tumour tissue with several steroids, evidence was obtained that the estradiol binding was inhibited, in a molar concentration ratio not far beyond the physiological range, by 5-androstene-3/&17/l-diol, a steroid closely related to DHEA. If these in vitro findings may be applied to in uiuo conditions, it is conceivable that androstenediol is a regulating agent of estrogenic action at the cellular level.

Abstract.A solid-phase extraction procedure for estradiol-17β from bovine blood plasma with a reversed phase cartridge was developed for the measurement of its concentration by radioimmunoassay. Estradiol-17β retained in the cartridge was eluted quantitatively with 70%( v / v ) methanol, although estrone was recovered in the same fraction. The apparent values measured by dextrancoated charcoal radioimmunoassay with an eluent of 75%( v / v ) methanol in the solid-phase extraction declined approximately by 10% as applied plasma volume was increased by 1 ml (r= 0.98, p<0.001). However, the values by double antibody radioimmunoassay were elevated approximately 8% per 1 ml plasma increase (r=0.82, p<0.001). The recovery of estradiol-17β added to bovine plasma in the extraction and radioimmunoassay with dextran-coated charcoal separation was approximately 75% under conditions of 2 ml plasma application, and concentrations calculated using buffer standards. Matrix effects on the assay recovery were negligible in the addition of solid-phase extract of castrated bull plasma to the standards. On the other hand, the average recoveries calculated with buffer and process standards in double antibody radioimmunoassay were 170% and 110%, respectively. The assay value in the solid-phase extraction showed good agreement with the value in the liquidliquid extraction and liquid chromatography after correction with the recovery rate (r=0.99). In conclusion, solid-phase extraction with a reversed phase cartridge is a practical extraction method for the measurement of estradiol-17β during the estrous cycle in cattle.
