Radioimmunoassay of Plasma Adrenocorticotrophin in Cushing's Syndrome

Abstract: The present study describes a radioimmunoassay for plasma adrenocorticotrophin (ACTH) in man with a sensitivity of at least 15 pg/ml.In-vitro studies using various fragments of ACTH showed immunobiological dissociation. However, this phenomenon does not appear to be of major importance in vivo, since stimulation and suppression tests showed overall parallel changes of immunoreactive plasma ACTH and plasma cortisol. The results obtained from patients with various forms of Cushing's syndrome are presented. It is… Show more

“…Sufficient pituitary response of ACTH to LVP in Cushing's disease and no response of ACTH to LVP in adrenal tumors were demonstrated. These findings were already reported in the previous literature (Ney et al, 1963;Graber et al, 1965;Demura et al, 1966;Demura et al, 1972;Croughs et al, 1973) In patients with totally adrenalectomized Cushing's disease, the infusion of 150mg hydrocortisone did not suppress plasma ACTH by 30 min, while the suppression was already observed by 30min in 3 patients with Addison's disease whose basal ACTH levels were also high. This finding suggests the abnormality in the feedback mechanism of ACTH secretion continuing after the clinically effective treatment.…”

“…We checked the antiserum against similar fragments and obtained similar results (Fig. 1), with the exception that the slope of the percentage bound v. log-dose curve of a25-39 ACTH was much flatter than that found by Croughs et al (1973). The slopes of serial dilutions of rat pituitary extracts and incubation medium from flasks containing rat pituitaries (in-vitro assay for CR-activity; below) were similar to that of the standard (Fig.…”

“…Blood levels of ACTH were measured, using an immunoassay method (Croughs, Tops & de Jong, 1973) in 40 patients during the last 2 weeks of pregnancy. Subjects were chosen who were free of serious medical disease and who had no apparent obstetrical disorder.…”
