Radioautographic analysis of 3H‐fucose utilization by fibroblasts of the periodontal ligament

Abstract: The secretion of 3labeld glycoproteins from periodontal ligament fibroblasts was, studied by light and electron microseopic radioautography. At 5, 10. and 20 minutes after intravenous injection, 3H‐fucose was concentrated in Golgi cisternae and saccules. By 35 minutes, the label was dispersed to the cell periphery and extracellular matrix. At 8 hours after injection, almost all of the radioactive label was associated with the cell surface or the adjacent extracellular matrix. Labeled glycoprotein was contained… Show more

“…Free 'H-fucose, labeled metabolie glyeoprotein intermediates, and procollagen are readily soluble and are assumed to be washed frotn tissues during fixation and processing (Bennett, Leblotid & Haddad 1974), Keratan sulfate is not a major component of the rat transseptal ligament (Johnson 1988). Cho & Garant (1986) describe the incorporation of 'H-fucose into giycoproteins of the periodontal ligament by fibroblasts. It is probable that fibroblasts of the periodontal and transseptal ligaments utilize 'H-fucose similarly.…”

“…It is probable that fibroblasts of the periodontal and transseptal ligaments utilize 'H-fucose similarly. The Golgi becomes labeled within 5-10 min of isotope injection, secretion granules within 20 min, and extracellular matrix within 35 min (Cho & Garant 1986). In the present study, significant tissue labeling was observed 1 h after isotope injection, confirming the observations of Cho & Garant (1986).…”
