R132H IDH1 sensitizes glioma to the antiproliferative and cytotoxic effects of BET inhibition

Abstract: IntroductionMutations in isocitrate dehydrogenase 1/2 (IDHmut) identify a subset of gliomas that exhibit epigenetic dysregulation via aberrant DNA methylation. These tumors are ultimately fatal and lack effective therapeutic strategies. Considering the epigenetic dysregulation of IDHmut gliomas, we hypothesized that epigenetic-targeting drugs may yield therapeutic benefits in gliomas bearing IDHmut. One set of targets includes the Bromodomain and Extraterminal (BET) family of transcriptional coactivators.Metho… Show more

“…In current clinical practice, the authoritative guidelines recommend genetic molecular biological markers, particularly the isocitrate dehydrogenase (IDH-1 ) gene, for glioma detection (Haddock et al, 2022;Wang et al, 2022). Single nucleotide polymorphisms (SNPs) of IDH-1 , especially mutations in motif 132, were found in more than 80% of gliomas and played a vital role in diagnosis, classification and prognosis in gliomas (Nabors et al, 2017;Sears and Woolard, 2022). Thus, the accurate and effective SNPs of IDH-1 gene detection make glioma therapy more promising.…”
