Quinine localizes to a non-acidic compartment within the food vacuole of the malaria parasite Plasmodium falciparum

Abstract: BackgroundThe naturally fluorescent compound quinine has long been used to treat malaria infections. Although some evidence suggests that quinine acts in the parasite food vacuole, the mechanism of action of quinine has not yet been resolved. The Plasmodium falciparum multidrug resistance (pfmdr1) gene encodes a food vacuolar membrane transporter and has been linked with parasite resistance to quinine. The effect of multiple pfmdr1 copies on the subcellular localization of quinine was explored.MethodsFluoresce… Show more

“…In order to analyze whether PfRAB7-and PfATG8-positive vesicles were acidified-a hallmark for the formation of autolysosomes-nonstarved and amino acid-deprived parasites were incubated with the acidotropic dye LysoTracker Red-DND99, which has been reported to label the acidic food vacuole within the parasite. 21 Colocalization between PfRAB7 and LysoTracker Red-DND99 was evident to a certain degree even under normal conditions (Fig. 4A, upper panel, Pearson's coefficient from analysis of 13 images, r = 0.56 ± 0.04), which is consistent with PfRAB7 being present in late endosomes, whereas the colocalization between PfATG8 and LysoTracker Red-DND99 was low under the same conditions (Fig.…”

“…To further examine whether these DV structures are physiologically intact, we stained live gametocytes with LysoTracker, a lysosomotropic dye that has been used to mark this highly acidic compartment in P. falciparum asexual blood stages (Bohorquez et al, 2012). Lyso-Tracker fluorescence was observed in a defined area that co-localized with hemozoin granules in all stages of gametocyte development (Fig.…”

“…Confirmation of the antimalarial activity of our library was done by performing 48-h IC 50 assays (Table 1). Although several of our test compounds are known to be fluorescent (11,12), the library exhibited negligible autofluorescence in our assay (data not shown). In the first round of screening, parasites were treated with the test compounds at 10 M and assayed with the ImageStream, a flow cytometer with imaging capabilities.…”
