Quercetin nanoparticle complex attenuated diabetic nephropathy via regulating the expression level of ICAM-1 on endothelium

Abstract: The purpose of the study was to reveal the therapeutic effect of quercetin (QUE) nanoparticle complex on diabetic nephropathy (DN) by regulating the expression of intercellular adhesion molecular-1 (ICAM-1) on endothelium as compared to free QUE. QUE 10 mg/kg as a single abdominal subcutaneous injection daily for 8 weeks continuously in diabetic rats and 10 mg/kg QUE nanoparticle complex as a single abdominal subcutaneous injection every 5 days, continuously administered for 8 weeks to diabetic rats. Blood and… Show more

“…Despite emerging clinical candidates under investigation for the treatment of DN, there is a lack of successful new treatments for DN. Some innovative treatment make use of traditional anti-diabetic drugs (insulin and exenatide), cytokines, metabolic modulation, nucleotide and quercetin 16 . Quercetin is an active compound for anti-cancer, anti-bacterial, anti-allergic and analgesic, and improves DN through various signaling pathways, making it a promising compound for the management of these diseases 7,19 .…”

“…In this regard, nanomedicine, especially gold nanostructure, has recently been used for the treatment of diabetic complications due to its higher bioavailability and its modulatory potential on hyperglycemia-induced redox-regulated cellular signaling mechanism. 36 The methanolic fraction of PPE was used as a reducing agent as well as a stabilizer in the green synthesis of AuNP. The phenolic compounds of PPE, especially ellagic acid and its derivatives, reduced Au 3+ to Au 0 and stabilized the AuNP through surface interactions with C=O and −OH groups of phenols.…”

“…51 Flavonoids, for instance, have been shown to attenuate DN more efficiently than commercial antidiabetic drugs via suppressing the activation of NF-κB and decreasing ICAM-1 and iNOS. 52,53 As shown here, UPARANT inhibits up-regulated levels of NF-κB, which, in addition to activate gene transcription of inflammatory factors, 54 promotes uPA transcription 55 and regulates ECM production. 56 In addition, NF-κB regulation of ICAM-1 positively correlates with nephropathy 57 by influencing mesangial cell proliferation.…”

“…This is particularly interesting since inflammation has been shown to play a major role in diabetic kidney disease and the suboptimal efficacy of the current therapeutic strategies may depend on their limited impact on inflammatory processes . Flavonoids, for instance, have been shown to attenuate DN more efficiently than commercial antidiabetic drugs via suppressing the activation of NF‐κB and decreasing ICAM‐1 and iNOS . As shown here, UPARANT inhibits up‐regulated levels of NF‐κB, which, in addition to activate gene transcription of inflammatory factors, promotes uPA transcription and regulates ECM production .…”
