Quercetin‐Induced miR‐369‐3p Suppresses Chronic Inflammatory Response Targeting C/EBP‐β

Abstract: Scope Dendritic cells (DCs) are the most potent antigen‐presenting cells that play an important role in the crosstalk between the innate and the adaptive immune response. Quercetin exposure is identified as an effective strategy to suppress the inflammatory response induced by LPS. Methods and results In this study, using a next‐generation sequencing analysis, the effect of quercetin on microRNAs (miRNAs) expression in DCs is examined. A signature of 113 miRNAs that are differentially regulated in LPS‐stimulat… Show more

“…In our recent study, we identified a specific signature of 113 miRNAs that were differentially regulated in LPS-stimulated BMDCs after quercetin exposure, along with their candidate target genes. Among these miRNAs, we mechanistically demonstrated that the induction of miR-369-3p is able to downregulate the inflammatory response induced by LPS, mimicking the effect of quercetin exposure 11 .…”

“…Que may reduce infection ( Li et al, 2016 ), hepatic lipemic-oxidative damage ( Cui et al, 2013 ) ( Zhang et al, 2016b ), and antioxidant risk ( Xu et al, 2019 ). In addition, Que is known to exert a modulating action on immunity ( Galleggiante et al, 2019 ). As regards its mechanism of action, in some cell lines Que was able to inhibit the production of TNF in macrophages ( Tang et al, 2019 ), IL-8 in A549 lung cells induced by lipopolysaccharide (LPS) ( Geraets et al, 2007 ), and TNF-α and IL-1α mRNA levels in glial cells, causing a decrease in neuronal cell death induced by microglial activation ( Li et al, 2016 ).…”

“…Furthermore, they showed that the expression of pro-inflammatory cytokines is positively regulated by C/EBP-β and is a direct target of miR-223 [91]. Moreover, in bone marrow-derived DCs and intestinal DCs, the regulation of C/EBP-β by miR-369-3p suppresses the chronic inflammatory response [100].…”
