Quantitative proteomics analysis reveals glutamine deprivation activates fatty acid β-oxidation pathway in HepG2 cells

Abstract: Glutamine, a multifunctional amino acid, functions in nutrient metabolism, energy balance, apoptosis, and cell proliferation. Lipid is an important nutrient and controls a broad range of physiological processes. Previous studies have demonstrated that glutamine can affect lipolysis and lipogenesis, but the effect of glutamine on the detailed lipid metabolism remains incompletely understood. Here, we applied the quantitative proteomics approach to estimate the relative abundance of proteins in HepG2 cells treat… Show more

“…The normalization of weight was regulated by increased energy expenditure, but not by caloric restriction or increased activity, and a transient induction of the energy balance regulating hormone fibroblast growth factor 21 (FGF21) play a critical role in this procession [ 30 ]. Glutamine as Leu metabolites plays a crucial role in various cellular processes, such as energy balance, apoptosis, and cell proliferation; it could activate the fatty acid β-oxidation pathway in HepG2 cells [ 31 ]. This result shows that glutamine deprivation can activate the fatty acid β-oxidation pathway to regulate lipid metabolism.…”

“…Glutamine plays a crucial role in various cellular processes, such as in energy balance, apoptosis, and cell proliferation and, its deprivation can activate the fatty acid β-oxidation pathway [150,151]. For instance, there has been a controversy linking glutamine with the prediction of the T2D.…”

“…Previously, using the iTRAQ-based approach, we have characterized the proteome of HepG2 cells treated with either Gln deprivation or Arg supplementation 19, 20 . However, to our knowledge, there is no literature reporting proteome scale alterations induced by Leu deprivation in liver cells.…”
