Quantitative Expression of Toll-Like Receptors TLR-7 and TLR-9 on Peripheral Blood Mononuclear Cells in Leukemias

Abstract: Background: Toll-like receptors (TLRs) are agents of innate and adaptive immunity, involved in tumor activation. The expression and functionality of TLR on leukemic cells have seldom been investigated. The aims of the study were to throw some light on the quantitative expression of TLR-7 and TLR-9 on leukemic cells and to compare it with their expression on mononuclear cells of healthy volunteers.

“…In this study, the TLR9 was ranging from 0.10 to 2.40% with a median of 1.30%. This finding coinciding with that reported by Morsi et al [16], where they found a range of (0.941-1.571%) with a mean of 1.35% among 15 AML patients.…”

“…In addition, in most studies, TLR6 C > T rs5743810 is associated with infectious diseases like tuberculosis in the African population and the induction of resistance to asthma in children 35 . In CLL, high TLR6 expression was observed in cells of the patients 36 , but it is still not clear what the role of this polymorphism is in leukemia.…”

“…In AML, the [C] allele of TLR9 C > T rs187084 and [T] TLR9 C > T rs5743836 are associated with disease development, as is the TT genotype of the TLR9 C > T rs5743836 polymorphism with relapse episodes 40 . Additionally, the TLR9 gene is significantly expressed in chronic lymphocytic leukemia cells 36 .…”

“…In other hematological cancers, such as acute myeloid leukemia (AML), the mRNA expression levels of TLR2 and TLR4, but not of TLR9, were increased, contrary to what was observed in ALL [ 37 ], and expression of TLR7 and 9 was decreased in chronic myeloid leukemia (CML) [ 38 ]. In chronic lymphocytic leukemia (CLL), there was an increase in the expression of TLRs 1, 2, 6, 7, 9, and 10 [ 38 , 39 ].…”
