Quantitative Colocalization Analysis of Multicolor Confocal Immunofluorescence Microscopy Images: Pushing Pixels to Explore Biological Phenomena

Abstract: Quantitative colocalization analysis is an advanced digital imaging tool to examine antigens of interest in immunofluorescence images obtained using confocal microscopes. It employs specialized algorithms to estimate the degree of overlap of fluorescence signals and thus enables acquiring important new information not otherwise obtainable using qualitative approaches alone. As raw confocal images have high levels of background, they should be prepared to become suitable for reliable calculation of colocalizati… Show more

“…Briefly, separate volumes were defined for LROs (based on Lamp staining) and GrB by automatic thresholding based on absolute intensity. Overlap coefficients according to Manders, 51 which range from 0 (no colocalisation) to 1 (100% colocalisation) and represent the true degree of colocalisation, were determined.…”

“…2E). Using Manders' colocalization coefficiency (21), above and below the endothelium, revealed a significant decrease in the degree of overlap specifically within the transmigrated part of the cell (Fig. 2F).…”

“…Images were acquired with a confocal microscope (Olympus, Japan). To determine the colocalization level of A 1 R and EGFR, Pearson's and Manders' coefficients were calculated using FV10-ASW2.1 software (Olympus) according to the previously reported analysis methods [23] . Briefly, Pearson's correlation coefficient describes the correlation of the intensity distribution between the channels, ranging from -1.0 to 1.0, where 0 indicates no significant correlation.…”
