Quantitative Assessment of the Chloroplast Lipidome

Abstract: In plants and algae, photosynthetic membranes have a unique lipid composition. They differ from all other cellular membranes by their very low amount of phospholipids, besides some phosphatidylglycerol (PG), and high proportion of glycolipids. These glycolipids are the uncharged galactolipids, i.e., monogalactosyldiacylglycerol and digalactosyldiacylglycerol (MGDG and DGDG), and an anionic sulfolipid, i.e., sulfoquinovosyldiacylglycerol (SQDG). In all photosynthetic membranes analyzed to date, from cyanobacter… Show more

“…In these conditions, the produced ions were mainly present as H - , H + , NH 4 + , or Na + adducts. Lipids were identified by MS analysis with their precursor ion or by neutral loss analyses after low energy collision-induced dissociation, as previously described (Gros and Jouhet, 2018).…”

“…Lipid classes were separated and their FA composition was analysed as described by Gros and Jouhet, 2018, with some minor differences. Shortly, after dilution of lipid extract in 1 mL of chloroform, a 50 mL sample was methylated (1 h, 100°C) and the total fatty acids were quantified by GC-FID.…”

“…The volume corresponding to 750 nmol of FA was loaded in pre-activated Thin Layer Chromatography (TLC) silica plates (105721, Supelco, Germany) and different plates were used to separate polar and neutral lipids, respectively. Migration and identification of lipid classes in TLC plates were done accordingly to Gros and Jouhet, 2018 with the difference that a solution of primuline 0.5% v/v in 8:2 acetone: deionised water was employed as a staining agent. After identification, the silica from the spots was scrapped and collected into separate glass tubes.…”

“…The individual spots corresponding to di erent lipid classes were scratched from the TLC plate and collected separately in a fresh glass tube. Each sample were methylated separately as mentioned above and analysed on GC to obtain the fatty acid pro les of each lipid class [385,386].…”
