Quantitative assessment of brown adipose tissue metabolic activity and volume using 18F-FDG PET/CT and β3-adrenergic receptor activation

Abstract: BackgroundBrown adipose tissue [BAT] metabolism in vivo is vital for the development of novel strategies in combating obesity and diabetes. Currently, BAT is activated at low temperatures and measured using 2-deoxy-2-18F-fluoro-D-glucose [18F-FDG] positron-emission tomography [PET]. We report the use of β3-adrenergic receptor-mediated activation of BAT at ambient temperatures using (R, R)-5-[2-[2,3-(3-chlorphenyl)-2-hydroxyethyl-amino]propyl]-1,3-benzodioxole-2,2-dicarboxylate, disodium salt [CL316,243] (a sel… Show more

“…The results of this study show that 18 F-FDG uptake of interscapular BAT is prominently increased by a single challenge with CL316,243, which is consistent with acute metabolic responses previously ob- served in mice (17,18) and humans (5,6). Interscapular BAT thus served as a positive control in our study to investigate the ability of 18 F-FDG imaging to monitor beige cell recruitment into WAT.…”

“…In identifying the mediators for increased 18 F-FDG uptake by b3AR stimulation, BAT displayed high expression of both GLUT4 and hexokinase-II regardless of treatment (Fig. 3).…”
