Quantitative Analysis of Herpesvirus Sequences from Normal Tissue and Fibropapillomas of Marine Turtles with Real-Time PCR

Abstract: Quantitative real-time PCR has been used to measure fibropapilloma-associated turtle herpesvirus (FPTHV) pol DNA loads in fibropapillomas, fibromas, and uninvolved tissues of green, loggerhead, and olive ridley turtles from Hawaii, Florida, Costa Rica, Australia, Mexico, and the West Indies. The viral DNA loads from tumors obtained from terminal animals were relatively homogeneous (range 2-20 copies/cell), whereas DNA copy numbers from biopsied tumors and skin of otherwise healthy turtles displayed a wide vari… Show more

“…Further work on the evolution of (Ene et al, 2005;Greenblatt et al, 2005;Quackenbush et al, 2001). Amplifications were performed in a total volume of 8 ml, with 1 ml lesion genomic DNA (including both target and green turtle DNA) at a concentration of~10 ng ml 21 , 4 ml Fermentas DreamTaq PCR Master Mix or 5Prime Master Mix, 2.2 ml MilliQ water and 0.4 ml each primer at 10 mM.…”

“…Fibropapillomatosis is a major cause of stranding in Hawaiian green turtles (Chaloupka et al, 2008b) and is associated with an alphaherpesvirus, but the role of this virus in disease causation remains unknown (Quackenbush et al, 2001;Lackovich et al, 1999;Greenblatt et al, 2005). Interestingly, FP in Hawaiian green turtles was known long before the Palaau outbreak but was rare (Herbst, 1994).…”

“…It has gained wide acceptance as a diagnostic tool due to its speed, sensitivity, reproducibility and the reduced risk of carry-over contamination (Mackay et al, 2002). This technique has been employed in diagnos ing and quantifying viruses infecting humans (Jabs et al, 2001;Kearns et al, 2001;Komurian-Pradel et al, 2001) and animals (Quackenbush et al, 2001;Smith et al, 2001;Markowski-Grimsrud et al, 2002).…”
