Quantified postsurgical small cell size CTCs and EpCAM+ circulating tumor stem cells with cytogenetic abnormalities in hepatocellular carcinoma patients determine cancer relapse

Abstract: Detection of hepatocellular carcinoma circulating tumor cells performed with conventional strategies, is significantly limited due to inherently heterogeneous and dynamic expression of EpCAM, as well as degradation of cytokeratins during epithelial-to-mesenchymal transition, which inevitably lead to non-negligible false negative detection of such "uncapturable and invisible" CTCs. A novel SE-iFISH strategy, improved for detection of HCC CTCs in this study, was applied to comprehensively detect, in situ phenoty… Show more

“…At present, the application of CTCs in liver cancer is basically limited to staging, prognosis 40 and therapeutic response evaluation. [41][42][43][44] Protein markers are classic tumuor markers with known stability. In general, good protein markers have considerable advantages in sensitivity and specificity.…”

“…3A. The clinical relevance of different types of aneuploid CTCs has been demonstrated, for example, highploid CTCs has been shown to be associated with tumor resistance and relapse [9,10]. However, the clinical significance of aneuploid CTECs are still unclear.…”

“…Stem cell-like CTCs, which are called circulating tumor stem cells (CTSCs), express putative stem cell markers, such as aldehyde dehydrogenase 1 (ALDH1) [8], DLG7 and BMI1 [9], CD44, and CD133 [10,11]. Moreover, epithelial cell adhesion molecule (EpCAM), which is an epithelial marker, has been detected in CTSCs with cytogenetic abnormalities [12]. Like CSCs, CTSCs go through a dynamic conversion upon exposure to environmental stress [13].…”
