Quantification ofAspergiullus flavus BFE96 by sybr green Q-PCR in black pepper

Abstract: A Real Time PCR system (Q-PCR) for the detection and quantification ofAspergillus flavus in black pepper has been used. Thenor1 gene, a gene of the aflatoxin biosynthetic pathway, served as target sequence. The determined copy numbers of thenor1 gene were compared to the cfu numbers obtained by plate counting. There was not a direct correlation of the results with both approaches, however the tendency was the same. In general both values increased with prolonged incubation time. The differences in copy numbers… Show more

“…The other approach is to enhance fungal cell wall disruption prior to DNA extraction. This was achieved by thoroughly mixing the sample material with alumina in a lysis buffer (4,16,20). As with the liquid nitrogen method, in some PCR assays additional bands appeared and the specific PCR products were rather weak (16,20).…”

“…Mayer et al (2003b) demonstrated that the tendency of nor-1 gene copies was the same as that of A. flavus CFUs in wheat with the prolonged incubation time (Mayer et al, 2003b). Additionally, nor-1 copies were demonstrated to be correlated to CFUs of A. flavus in pepper, maize, and paprika (Bagnara et al, 2000;Mayer et al, 2003a). Passone et al (2010) also developed an analytical method determining Aspergillus section Flavi based on nor-1 gene and demonstrated a good correlation (r = 0.613; P < 0.0001) between nor-1 copies and CFUs in naturally stored peanut.…”
