BackgroundThe Zika AIRS Project, a USAID-funded initiative worked across the Latin America and Caribbean regions from 2016 to 2019, as an emergency to contain the spread of the Zika virus. All entomological records in the target countries showed wide distribution and high abundance of Aedes aegypti populations, however the susceptibility profiles of these insects to insecticides commonly employed by vector control campaigns were in most cases incomplete or inexistent. In close collaboration with the Ministries of Health of individual countries, Zika-AIRS teams conducted insecticide susceptibility testing of an array of insecticides in A. aegypti populations of each country. Procedures applied met the standard international protocols instructed by the World Health Organization and Centers for Disease Control and Prevention.Methodology and main findingsThe insecticides tested were selected under categories such as pyrethroids, organophosphates and carbamate. Results showed A. aegypti populations displaying high and widely distributed resistance to all pyrethroids across countries, tolerance to organophosphates and full susceptibility to a carbamate. Key inconsistencies between testing methods are presented and discussed. Additionally, four kdr mutations were analyzed to detect molecular mechanisms of insecticide resistance. The screening for kdr mutations suggested the widespread nature of V1016I mutation, linked to pyrethroid resistance in A. aegypti populations distributed and sampled in the above mentioned regions.Conclusions and perspectivesThis multi-country study contributes with updated information to the public health decision-makers across Central America and the Caribbean. This study provided training and established technical networks for more effective and sustainable insecticide surveillance programs. Most but not all records of insecticide resistance in A. aegypti were consistent between methodologies, thus inconsistent issues are discussed here to call for further improvement in procedures and convey more practical guidelines for surveillance teams in countries where Aedes-borne diseases are endemic.Author summaryAt the forefront of the fight against arboviruses transmission is the insecticide-based vector control. All countries in the Latin American and Caribbean region invest valuable resources from their limited budget to acquire and implement insecticide-based tools, with non-existent or weak insecticide resistance monitoring programs. Hence, the USAID-funded Zika AIRS Project (ZAP) collaborated with the Ministries of Health of multiple countries to update the profile of susceptibility to insecticides in Aedes aegypti populations. We found widespread resistance to pyrethroid and organophosphate insecticides, which account to almost 100% of the products available to control adult mosquitoes. As we used both of World Health Organization and Centers for Disease Control and Prevention standard methods, we found many similarities and some inconsistencies in the susceptibility profiles obtained for the very same vector populations. Additionally, we obtained insight on potential molecular mechanisms of resistance across the countries, finding the kdr mutation V1016I possibly involved in loss of susceptibility.This study is the biggest cross-country update of insecticide resistance for Aedes aegypti in years, and it should be used as evidence for improving the selection of insecticides in these countries and a call for further support to maintain insecticide resistance monitoring programs.

Aedes aegypti, as one of the vectors transmitting several arboviruses, is a main target in mosquito control programs. Permethrin remains the major adulticide used to control these mosquitoes. The increasing percentage of permethrin resistant Aedes aegypti has become an important issue around the world. Knockdown resistance (kdr) is one of the major mechanisms related to permethrin resistance. On the other hand, detoxification genes including cytochrome P450 monooxygenases (P450) and glutathione S-transferases (GSTs) are also suggested as permethrin resistance apparatus. Here we selected a permethrin resistant (p-s) Aedes aegypti population from Florida and compared its mortality after exposure, median lethal dose (LD50), adult survivorship and larval development to several field populations. We used allele-specific PCR genotyping of the S989P, V1016I and F1534C sites in the sodium channel gene and gene expression analysis of several p450 and GSTs genes before and after permethrin exposure to determine their involvement in permethrin sensitivity between Ae. aegypti populations. Results indicated the p-s population had the highest resistance to permethrin based on LD50 and the mortality test. The larval development time did not significantly differ between the populations, however the p-s adults survived longer than the other populations. In the genotype study, p-s population had mostly homozygous mutations in all three mutant sites of the sodium channel gene. Detoxification gene expression studies showed that two p450 genes, AAEL009124 (CYP6N12) and, AAEL009127 (CYP6M11), were upregulated and, accession # AAEL006802, AAEL014891 (CYP6P12) and AAEL014619 (CYP9J22) were downregulated after 120 minutes of permethrin exposure in the p-s population. These results suggest that in highly permethrin resistant Aedes aegypti populations both kdr mutations and xenobiotic metabolism might be involved. Involvement of multiple mechanisms to achieve resistance to permethrin supports the need for implementing comprehensive mosquito control measures, such as an integrated pest management strategy, so that selection pressure for resistance is decreased without compromising control efforts while new methodologies are being developed.Author summaryPyrethroids have been applied as a major type of insecticide targeted at Aedes aegypti, a key vector in the transmission of several flaviviruses. Resistance to pyrethroids has emerged and has become a worldwide threat to mosquito control. Pyrethroid resistance usually occurs with knockdown resistance (kdr) where the voltage gated sodium channel is mutated. We selected a permethrin resistant (p-s) Aedes aegypti population from Florida and, along with two other field populations, examined three mutation sites, S989P, V1016I and F1534C. The data showed the p-s population had the most homozygous mutations which correlated to the permethrin resistance level. Besides kdr, detoxification genes also have been identified to have pyrethroid metabolizing abilities. We found two cytochrome P450 monooxygenases genes, CYP6N12 and CYP6M11, were overexpressed in the p-s population after permethrin exposure, suggesting a role in resistance to permethrin. Together, our results provide information about potential mechanisms used in major vectors with high insecticide resistance.

Insecticide resistance in Aedes aegypti: An impact from human urbanization?

In the city of Magelang, Indonesia, the distribution of Dengue Haemorhagic Fever (DHF) cases tend to be clustered, ever changing along with human urbanization from 2014 to 2017. Although DHF cases have been less reported in the city of Magelang for the past 5 years, vector control measures by using insecticide space spraying, particularly permethrin, have been continuously performed. Current study aimed to detect kdr mutations associated with pyrethroid resistance in Ae . aegypti and to study possible association between insecticide resistance and DHF case distribution related to human urbanization. The study was a cross sectional study conducted in 3 sub-districts in the city of Magelang, Central Java, Indonesia. Eggs of Ae . aegypti collected from 195 sample households were reared and were tested for resistance to pyrethroids by using PCR. Primers AaSCF1 and AaSCR4, and primers AaSCF7 and AaSCR7 were used in detecting presence of mutation in VGSC IIS6 and IIIS6 gene, respectively. Fragments of amplified DNA were sequenced and were analyzed. Spatio-temporal using Standard Deviational Ellipse (SDE) was performed to obtain mapping of DHF case distribution trends. The total number of DHF case was 380 cases, with the most cases (158) occurred in 2015 and the least cases (66) reported in 2017. DHF case distribution was grouped into several clusters. SDE calculation demonstrated movement of DHF case in the direction to principal arterial road, suggesting link to urbanization. Gene sequencing demonstrated VGSC IIS6 gene mutation (S989P and V1016G) in Ae . aegypti collected from study areas, indicating resistance to permethrin. VGSC IIIS6 gene mutation was not found. Current study concluded that multiple kdr mutations associated with resistance to pyrethroid was detected in Ae . aegypti , and that human urbanization may have a role in the development of such resistance.
