Quantification of ocular inflammation: Evaluation of polymorphonuclear leucocyte infiltration by measuring myeloperoxidase activity

Abstract: Myeloperoxidase (MPO) activity was measured in rabbit cornea and iris-ciliary body to quantitate the infiltration and accumulation of polymorphonuclear leucocytes (PMN's) following an inflammatory stimulus. Following injection of clove oil into the cornea, MPO activity could be detected in the cornea at 6 hr, reaching a maximum at 12 hr, and falling to non-detectable levels at 72 hr. MPO activity was only detected in the iris-ciliary body 24 hr after intracorneal clove oil injection. MPO activity in the iris-c… Show more

“…The slope of the line was determined for the individual samples and used to calculate the number of units of MPO in the tissue. One unit of MPO activity is defined as that degrading 1 mol of peroxide per minute (23). Results are reported as units of MPO/cornea.…”

“…The change in absorbance at 460 nm was monitored for 5 min at 30-s intervals, and the results were expressed as units of MPO per cornea. One unit of MPO activity is equivalent to 2 ϫ 10 5 PMN (24).…”

“…The slope of the line was determined for each individual sample and used to calculate the number of units of MPO in the tissue. One unit of MPO activity is defined as that degrading 1 mol of peroxide per minute (21). To ensure reproducibility of the data, the MPO assay was repeated once similarly.…”
