Quantification of Network Perfusion in ASL Cerebral Blood Flow Data with Seed Based and ICA Approaches

Abstract: Independent component analysis (ICA) or seed based approaches (SBA) in functional magnetic resonance imaging blood oxygenation level dependent (BOLD) data became widely applied tools to identify functionally connected, large scale brain networks. Differences between task conditions as well as specific alterations of the networks in patients as compared to healthy controls were reported. However, BOLD lacks the possibility of quantifying absolute network metabolic activity, which is of particular interest in th… Show more

“…Absolute CBF values obtained using ASL in the whole brain and specific brain regions have been shown to be reproducible across time scales of minutes, hours to days (Chen et al, 2011; Jain et al, 2012; Jann et al, 2013; Wu et al, 2011). There is a good correlation between ASL CBF and the gold standard of 15 O-PET in both resting state and activation studies (Feng et al, 2004; Kilroy et al, 2013; Ye et al, 2000).…”

“…There is a good correlation between ASL CBF and the gold standard of 15 O-PET in both resting state and activation studies (Feng et al, 2004; Kilroy et al, 2013; Ye et al, 2000). Besides providing a robust mean CBF averaged across a few minute scan, it has been shown to be feasible to perform dynamic FC analysis of the ASL perfusion image series (Chuang et al, 2008; Dai et al, 2013; Jann et al, 2013; Liang et al, 2012; Zou et al, 2009), similar to BOLD FC analysis. To date, however, very few studies have systematically addressed the similarity of RBNs detected using BOLD and ASL contrasts, as well as their reliability across sessions and scanners.…”

“…For instance, due to the relatively low temporal resolution of ASL scans, perfusion time series from multiple subjects are often concatenated, for instance, in a group ICA (Jann et al, 2015a;Jann et al, 2013). In addition, as CBF and BOLD provide complementary information, it is appealing to combine ASL and BOLD for characterizing spatiotemporal and quantitative properties of specific brain networks, as proposed by a few recent studies (Jann et al, 2013;Zhu et al, 2013). Furthermore, joint CBF-and BOLD-based FC analyses offer mutual validation of findings using each modality, thereby improving confidence for statistical inferences.…”

“…6 In addition, it offers a more direct approach to measure neural activity than BOLD due to the close coupling of neural activity and CBF. 6,10 ASL has demonstrated its ability to detect resting state functional networks, [11][12][13][14][15][16][17] first in a sensorimotor network using seed-based analysis and later mapping large-scale networks using independent component analysis (ICA). Our ASL preliminary resting state studies have also been reported in abstract forms.…”
