In hens, the granulosa layer is the primary source of anti-Mullerian hormone (AMH), as it is in mammals. Small follicles express the greatest amount of Amh mRNA with less in the larger follicles. Laying hens have a distinct ovarian hierarchy of follicles while broiler breeder hens often have excessive follicle growth with a disrupted hierarchy. The objective of Experiment 1 was to examine Amh expression in two strains of hens differing in ovulatory efficiency. Amh expression was greater (P!0.01) in broiler breeder hens (nZ6) as compared with laying hens (nZ6). Experiment 2 was designed to examine whether alterations in follicular development due to diet, within the broiler breeder hens, were correlated with changes in the expression of Amh. Restricted feeding (RF) in broiler breeder hens promotes optimal follicular development. Egg production in broiler breeder hens on full feed (FF; nZ8) was 78% that of hens on RF (nZ9). The number of large follicles (P!0.05), total ovarian weight (P!0.01), and Amh mRNA expression were greater in FF hens as compared with RF hens (P!0.01). There was no difference in FSH receptor expression between the two groups. A direct nutritional effect was not supported because culture of granulosa cells with varying concentrations of glucose and insulin showed no effect on granulosa Amh expression. Finally, testis-conditioned medium resulted in a dose-related increase in granulosa cell proliferation, which could be inhibited by preincubation with AMH antibody. AMH may enhance granulosa cell proliferation through an autocrine or paracrine mechanism although excessive AMH may inhibit optimal follicle selection.

The implantation of two testes from 13-day-old male chick donor embryos into the extra-embryonic celom of 3-day-old female embryos induces the masculinization of their ovaries up to a total and definitive inversion of their gonadal sex, i.e., the differentiation of testes in the female hosts. Pretreatment of the donors with estradiol (E2) between day 11 and 13 counteracts the testis-inducing activity of the implants, while co-treatment of donors with both tamoxifen (TAM) and E2 at the same stage restores the initially observed activity. The treatment of 3-day-old male donor embryos with E2 causes the differentiation of their left gonad into an ovotestis totally devoid of testis-inducing activity once grafted in the same conditions as above. An additional treatment with TAM of the grafted host embryos does not modify the results obtained when E2-treated male gonads are grafted to normal host embryos. This shows that the lack of testis-inducing activity exhibited by the E2-treated grafts can not be attributed to a protecting action of endogenous estrogens on the gonads of the host. On account of previous work showing the inhibition by E2 of the Müllero-regressive activity of the chick embryonic testis, our present results can be interpreted in terms of E2-down regulation of Anti-Müllerian Hormone (AMH or MIS), which appears to be a good candidate as testis-inducer. The relevance of our results to the phenomenon of gonad differentiation is discussed.

Müllerian ducts of male chickens undergo regression around day 12 of incubation, but the underlining mechanisms remain unclear. The purpose of this study was to identify factors that contribute to regression of the Müllerian duct in the chicken. We first employed annealing control primer-based RT-PCR to screen candidate genes differentially expressed in the Müllerian ducts between male and female. Four differentially expressed genes (MSX2, GAL10, VCP and PLCH1) were partially sequenced. The expression of mRNA of the latter genes and MSX1 in the male and female Müllerian ducts were compared at 7.5, 8 and 9 days of incubation using semiquantitative RT-PCR. The results indicated that both MSX1 and MSX2 mRNA was highly expressed in the male Müllerian duct at day 9 of incubation, whereas, PLCH1 mRNA was lower in the male duct at day 9 of incubation compared to that of the female duct. Although VCP mRNA was expressed in both left and right female Müllerian ducts, no expression was detected in the male duct. Whole mount in situ hybridyzation analysis showed that the expression of MSX1 and MSX2 mRNA were localized specifically in the mesenchymal cells of the male Müllerian duct at day 9 of incubation. In contrast, VCP mRNA expression was observed in both mesenchymal and epithelial cells of the female Müllerian duct but not detected in the male duct. These results suggest that both up-regulation of MSX1 and MSX2 mRNA expression is involved in the regression of the Müllerian duct in male chicken embryo, whereas VCP expression is involved in development of the female duct.
