Quantification of miRNA-mRNA Interactions

Abstract: miRNAs are small RNA molecules (′ 22nt) that interact with their corresponding target mRNAs inhibiting the translation of the mRNA into proteins and cleaving the target mRNA. This second effect diminishes the overall expression of the target mRNA. Several miRNA-mRNA relationship databases have been deployed, most of them based on sequence complementarities. However, the number of false positives in these databases is large and they do not overlap completely. Recently, it has been proposed to combine expression… Show more

“…In contrast, miRNAs are transcriptionally regulated and in the majority of cases their expression levels directly correspond to their activity. In addition, unlike TFs that can serve as activators or repressors, miRNAs directly repress their targets, leading to anticorrelated expression levels between miRNAs and their targets (1,8,9,10,14). Finally, miRNAs often target fewer genes than TFs and their impact on each of their targets is limited (6,15).…”

“…This new particle-to-cell communication could assist the epigenetic reprogramming of protein expression patterns in target cells and thus, the identification of miRNA targets should represent a major goal for understanding their downstream effects and define their biological functions and key blocking events. In this respect, numerous computational tools are being developed for predicting and quantifying miRNA targets [71,72].…”
