“…For comparison, previously reported data (41) The reasons for the discrepancies between the findings from the above biochemical studies (42,43) and our studies are unknown at present. Although the anti-Ca2+ pump antibody (5F10) used in the previous immunological study (11) was produced against the human red cell membrane Ca2W pump, this antibody cross-reacts with the PMCA proteins of rat intestine (44), rat and human kidney (45,46), and human placenta (47); and, in our previous study (11), the 5F10 antibody immunohistochemically cross-reacted almost exclusively with a protein resident to the chicken intestinal basolateral membrane.…”

“…A direct effect on Ca2+ pump activities appears rather unlikely, for at least two reasons. Firstly, basal [Ca2+]i was identical in enterocytes from con-and def-animals indicating intracellular Ca2+ homeostasis including intact Ca2P pumps in both preparations, and secondly, it has been shown previously that Ca2+ pump capacities in basolateral membranes of porcine duodenum are not influenced by calcitriol deficiency (Timmermans et al 1991). An increased Ca2+ pump activity due to elevated [Ca2+]i in the deficient animals should be excluded since active transcellular Ca2+ transport was completely absent in duodenal tissues (Kaune et al 1992).…”

“…Purification was monitored by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (Laemmli, 1970) and fluorescence spectrometry (excitation, 276 nm; emission, 306 nm) (Bryant & Andrews, 1983). The competitive ELISA procedure has been described elsewhere (Timmermans, Kaune, Bindels & Van Os, 1991). Calbindin-Dgk concentrations are expressed as microgram calbindin-D9k per milligram of soluble protein, with 78 4 + 3 1 mg soluble protein (mean + S.D.…”

“…Basolateral efflux of Ca 2+ occurs against a considerable electrochemical gradient, which is mainly mediated via Ca 2+ -ATPase activity ("plasma membrane Ca 2+ -ATPase", PMCA1b; Sepulveda and Mata, 2004), with only a minor contribution of the Na + /Ca 2+ exchange (NCX) mechanism at least in pigs (Timmermans et al, 1991;Kaune, 1996;Hoenderop et al, 2000). Controversy still exists about the influence of calcitriol on basolateral Ca 2+ efflux.…”
