Quality of bull spermatozoa after preparation by single-layer centrifugation

Abstract: The present study aimed to evaluate the effect of single-layer centrifugation (SLC) through a species-specific colloid (Androcoll-B; patent pending, J. M. Morrell) on bull sperm quality. Computer-assisted sperm analysis of motility and flow cytometric analysis of sperm viability (SYBR-14/propidium iodide staining), chromatin integrity (acridine orange staining), reactive oxygen species production [Hoechst 33258-hydroethidine-2',7'-dichlorodihydrofluorescein diacetate (HO-HE-DCFDA) staining], mitochondrial memb… Show more

“…Nevertheless, it must be highlighted that DNA fragmentation observed was very low and within the normal range described previously in other studies (1.5-5.0%) [34][35][36]. This result contrasts significantly with previous studies using other sperm pretreatments agents, such as DTT, which produced massive chromosome aberrations and high DNA fragmentation in mouse and bovine sperm [14,22].…”

“…Sperm plasma membrane integrity was assessed by flow cytometry after staining with SYBR®14 and propidium iodide (PI) (Goodla et al., ). Briefly, buffer B was added to aliquots of unselected and SLC‐selected samples to achieve a concentration of ~2 × 10 6 sperm cells/ml.…”

“…Previous studies with colloid centrifugation, using species -specific formulations of Androcoll, have reported beneficial effects in improving the motility, viability or mitochondrial activity of sperm samples, and removing dead cells, in red deer [10], boar [11], bull [12], or stallion [13], or buck [14]. In contrast, one of the main problems of the selection process with colloid centrifugation is the low yield in thawed samples.…”
