-Summary. Melilotus suaveolens Ledeb is a species of traditional medical plant for treating inflammationrelated disease. This study is to explore the inner anti-inflammatory mechanism on petroleum ether extract from Melilotus suaveolens Ledeb. Materials and Methods. Inflammatory cellular model was founded by intervention of lipopolysaccharide (LPS) on RAW264.7 cell line. Secretion of TNF-a, IL-1b, IL-6, NO and IL-10 in supernatant, mRNA expression of TNF-a, COX-2, iNOS and HO-1, protein expression of COX-2 and HO-1, activation of NFkB and ingredients in the extract were assayed. Results. The extract could not only reduce production of pro-inflammatory mediators by blocking NF-kB activation but promote release of anti-inflammatory mediator HO-1 significantly. The only active ingredient in the extract was coumarin and the concentration of coumarin in each 1 g extract was 0.27822 mg. Conclusion. Compared to Dexamethasone, the extract not only has similar effects on antagonizing proinflammatory mediators and cytokines but has effects on promoting production of anti-inflammatory mediators.

Buyang Huanwu Decoction (BYHYD) is a classical traditional Chinese medicinal prescription that has been widely used for treating cerebrovascular illnesses for hundreds of years. In this study, a comprehensive analytical method has been developed for quantitative analysis of the major constituents in BYHWD. This method was based on high-performance liquid chromatography coupled to a diode array and evaporative light scattering detectors (HPLC-DAD-ELSD) on a common reverse-phase C(18) column. HPLC coupled with on-line time-of-flight mass spectrometry (HPLC-TOF/MS) was additionally adopted to provide further validation for the constituents. It was found that 0.3% aqueous formic acid and acetonitrile was the optimum mobile phase for gradient elution. This method, which showed excellent precision and accuracy, was successfully applied to quantify the bioactive constituents in six BYHWD products. The validated HPLC-DAD-ELSD method, together with the HPLC-TOF/MS analysis, provided a new basis for assessing the quality of traditional Chinese medicinal prescription consisting of many bioactive components.

The emerging development of Extractive Reference Substance (ERS) is a methodology that meets the needs for quality control for Chinese Herbal Medicines (CHM) and respects the holistic viewpoint of Traditional Chinese Medicine (TCM) and its clinical use of multiple ingredients with synergistic effects. The convention of using just a selected few Chemical Reference Substances (CRS) cannot adequately assess the quality of intact CHM. A validated chemical spectrum of an ERS provides the global characteristics in order to more specifically identify and assess targeted CHM. This paper describes the fundamental concepts, potential significance, and basic criteria of ERS, along with methods of preparation and calibration. Given the diversity of CHM, the various problems that will occur in establishing the proper process of ERS will need to be solved in a step by step manner. The ERSs of Ziziphi spinosae semen and ERS of Fritillaria thunbergii bulbus are given as examples of the development of ERS and demonstrate why we are optimistic about the utility of this approach.
