Quality assessment on Polygoni Multiflori Caulis using HPLC/UV/MS combined with principle component analysis

Abstract: BackgroundPolygoni Multiflori Caulis, the dried caulis of Polygonum multiflorum Thunb., is one of the commonly used traditional Chinese medicines having antioxidant, anti-obesity, anti-inflammatory and antibacterial effects. Polygoni Multiflori Caulis used clinically or circulated on market have great differences in their diameters. However, to the best of our knowledge, no study has been reported on the qualities of Polygoni Multiflori Caulis with different diameters.ResultsSystematic HPLC/UV/MS chromatograph… Show more

“…The prototype components were identified or tentatively identified by comparing their retention times and TOF–MS data with those of authentic standards (Figure S3 and S4) or with the literature (Chen et al, , Huang et al, , Tang, Wang, Liu, Cheng, & Xiao, , Yang et al, , Zhao, Kao, et al, ). The known compounds were identified by comparing the accurate mass and retention time with authentic standards, and the unknown compounds were tentatively characterized by comparing the accurate mass and tandem mass spectrometric data with previous research findings.…”

“…To obtain a clearer view of the absorbed prototype components and metabolites profiles of TGG, the base-peak intensity chromatograms of high-drug-concentration plasma and bile samples were extracted in both positive and negative ion modes rather than a total ion chromatogram (Supporting Information Figures S1 and S2). The extracted ion chromatograms (EIC) of TOF-MS for the absorbed prototype components and metabolites of TGG are presented in Figures 1 and 2, which could avoid endogenous interference from complex biological matrices that may affect identification of the absorbed components and metabolites (Zhou et al, 2017), rather than base-peak intensity and total ion chromato- Table 1. The prototype components were identified or tentatively identified by comparing their retention times and TOF-MS data with those of authentic standards ( Figure S3 and S4) or with the literature (Chen et al, 2014, Huang et al, 2016, Tang, Wang, Liu, Cheng, & Xiao, 2016, Zhao, Kao, et al, 2013. The known compounds were identified by comparing the accurate mass and retention time with authentic standards, and the unknown compounds were tentatively characterized by comparing the accurate mass and tandem mass spectrometric data with previous research findings.…”

“…Recently, a number of chromatographic methods have been established to determine the major constituents of PMC through HPLC and LC–MS , but there is no study about comprehensive analysis of the chemical composition. Additionally, different pharmacological effects always result from variant chemical ingredients.…”
