“…Thus, Andrew and Morris (1965) demonstrated a net synthesis of alanine by Clostridium kluyvcri extract from uC02-acetyl coenzymc-A and hydrogen. Raeburn and Rabinowitz (1965) obtained a similar net synthesis in extracts of Clostridium acidi-urici. The source of re ducing power was reduced ferredoxin in both cases.…”

“…Brill et al (3) concluded that ferredoxin is involved in the oxidation of formate by extracts of M. omelianskii. Ferredoxin is also involved in the fermentation of pyruvate by several clostridia (17) as well as in its synthesis from acetyl-coenzyme A and C02 (1,15). Pyruvate seems to be involved in the total synthesis of acetate from CO2 (10), and it seems likely that pyruvate is a source of electron for this reduction of CO2 and that the reduction might occur via ferredoxin.…”

“…Shortly thereafter, we identified PFOR, designated pyruvate synthase, in a cell-free preparation from the purple sulfur bacterium Chromatium strain D (27), thereby demonstrating that Fdx could function directly in CO 2 fixation in a photosynthetic cell. Others soon reported the enzyme in clostridial species other than C. pasteurianum (2,115). However, despite this success, we were still faced with a major problem: Although we were able to reduce Fdx using H 2 /hydrogenase in Chromatium extracts, attempts to photoreduce Fdx with chlorophyllcontaining membrane preparations were unsuccessful, thus restricting the role of the reaction in photosynthesis.…”
