Pyruvate as a cell‐protective agent during cold atmospheric plasma treatment in vitro: Impact on basic research for selective killing of tumor cells

Abstract: Promising investigations on the selective killing of tumor cells were reported using cold atmospheric pressure plasma (CAP) in vitro. Pyruvate, a supplement in cell culture media, could protect cells against the action of reactive oxygen species. Here, we provide an overview of the in vitro studies concerning the selective killing of tumor cells by CAP with special emphasis on pyruvate concentration in the cell culture media. Several former studies were performed in media with different pyruvate content. Our c… Show more

“…This could be explained by the very low concentration of H 2 O 2 observed in the treated medium (< 1 µM). H 2 O 2 is likely rapidly scavenged by pyruvate in the culture medium [44], [45], [46], disabling it from having any long-term effect on the cells.…”

“…Consequently, all cell lines were treated in the same culture medium (DMEM with pyruvate) to avoid this bias. DMEM with pyruvate was chosen to highlight the influence of direct NTP treatment over the capacity of plasma to produce long-term RONS such as H 2 O 2 [66][67][68][69].…”

“…We incubated HaCaT cells in DMEM supplemented with 150 mg/L H 2 O 2 . We chose this concentration because, in previous work, we found an H 2 O 2 content of 120 mg/L for DMEM without pyruvate directly after CAP exposure and could show that the H 2 O 2 was depleted immediately by pyruvate in DMEM with 1 mM pyruvate [80]. In consideration of other anti-oxidative components contained in DMEM, such as Fe 3+ , which leads to the decomposition of H 2 O 2 by the Fenton reaction, we used the concentration of 150 mg/L.…”

“…The distance between the tip of the quartz capillary and the 96-well plate did not exceed 1 mm. In this way, the argon plasma had immediate contact with the media [45,80]. Argon gas treatment using the kINPen ® 09 without igniting the plasma for 60 s was used as a control (argon-activated medium, AAM).…”
