Синтез поліфтороалкілвмісних піролів як будівельних блоків для отримання фторовмісних алкалоїдів піролідинового ряду Мета. Вивчити синтетичні можливості циклізації N-(β-поліфтороацил)вінілпохідних проліну та N-заміщених гліцинів у поліфтороалкілпіроли, які є потенційними проміжними речовинами до фторовмісних алкалоїдів піролідинового ряду. Результати та їх обговорення. Синтезовано ряд N-(β-поліфтороацил)вінілпохідних проліну та N-заміщених гліцинів, які було трансформовано в поліфтороалкілвмісні піроли дією оцтового або трифтороцтового ангідриду. Експериментальна частина. Дією β-алкоксивініл-(поліфтороалкіл)кетонів на пролін та N-заміщені гліцини синтезовано N-(β-поліфтороацил)вінілпохідні відповідних амінокислот, які було трансформовано у поліфтороалкілвмісні піроли під дією оцтового або трифторооцтового ангідриду. Структуру і склад синтезованих речовин доведено даними 1 Н і 19 F ЯМР-спектроскопії та елементним аналізом. Висновки. Встановлено, що збільшення стеричних навантажень в N-(β-поліфтороацил)вінілпохідних проліну (подовження поліфтороалкільної групи з трифторометильної до перфтороетильної) та N-заміщених гліцинах (заміна N-метильної групи на N-бензильну) приводить до переважного утворення поліфтороацилпіролілацетатів при нагріванні в оцтовому ангідриді. Ряд поліфтороалкілвмісних піролів було синтезовано при дії трифторооцтового ангідриду на N-(β-поліфтороацил)вінілпохідні проліну та N-заміщені гліцини.

Background: The inherent glycosidase inhibitory activity and potentially therapeutic value of the polyhydroxylated pyrrolizidine alkaloids containing a hydroxymethyl substituent at the C-3 position have been well documented. Belonging to this class, the naturally occurring hyacinthacine C-type alkaloids are of general interest among iminosugar researchers. Their selective micromolar α -glycosidase inhibitory ranges (10 – 100 μM) suggest that these azasugars are potential leads for treating type II diabetes. However, the structures of hyacinthacine C1, C3 and C4 are insecure with hyacinthacine C5 being recently corrected. Objective: This review presents the hyacinthacine C-type alkaloids: their first discovery to the most recent advancements on the structures, biological activities and total synthesis. Conclusion: The hyacinthacine C-type alkaloids are of exponentially increasing interest and will undoubtedly continue to be reported as synthetic targets. They represent a challenging but rewarding synthetic feat for the community of those interested in accessing biologically active iminosugars. Since 2009, ten total syntheses have been employed towards accessing similarly related products but only three have assessed the glycosidase inhibitory activity of the final products. This suggests the need for an accessible and universal glycosidase inhibitory assay so to accurately determine the structure-activity relationship of how the hyacinthacine C-type alkaloids inhibit specific glycosidases. Confirming the correct structures of the hyacinthacine C-type alkaloids as well as accessing various analogues continues to strengthen the foundation towards a marketable treatment for type II diabetes and other glycosidase related illnesses.

Hepatic veno-occlusive disease (VOD) is a life-threatening complication of hematopoietic stem cell transplantation, which urgently requires effective prevention and treatment. Endothelial damage is recognized as the first event in patients with hepatic VOD. However, the mechanism by which endothelial injury induces thrombosis in hepatic VOD is still not clear. In the present study, monocrotaline (MCT) was used to induce endothelial cell injury in EA.hy926 cells to imitate in vitro hepatic VOD. MCT significantly increased apoptosis in EA.hy926 endothelial cells and the secretion of endothelial microparticles (EMPs) which can be used to reflect the level of endothelial injury. Additionally, MCT significantly enhanced the expression of soluble tissue factor (TF) and EMP-bound TF protein, suggesting that EMPs may participate in the development of hepatic VOD by regulating coagulation. Ginsenoside Rb1, a major constituent and effective ingredient of Panax ginseng, was found to significantly decrease MCT-induced endothelial injury and release of EMPs. Moreover, Ginsenoside Rb1 decreased soluble TF released by EA.hy926 cells and EMP-bound TF protein induced by MCT. These data suggest that ginsenoside Rb1 may serve as a potent prophylactic and/or as a treatment of hepatic VOD by protecting endothelial cells and preventing microthrombosis induced by endothelial injury.
