Pyroptosis, inflammasome, and gasdermins in tumor immunity

Abstract: The gasdermins (GSDM), a family of pore-forming proteins, consist of gasdermin A (GSDMA), gasdermin B (GSDMB), gasdermin C (GSDMC), gasdermin D (GSDMD), gasdermin E (GSDME) and DFNB59 (Pejvakin (PJVK)) in humans. These proteins play an important role in pyroptosis. Among them, GSDMD is the most extensively studied protein and is identified as the executioner of pyroptosis. Other family members have also been implicated in certain cancers. As a unique form of programmed cell death, pyroptosis is closely related… Show more

“…Additionally, it has been associated with drug resistance during tumor chemotherapy [34]. The GSDMS family plays a crucial role in pyroptotic processes, with GSDMB, GSDMC, GSDMD, and GSDME mediating enhanced immune responses against certain cancers [35]. Notably, GSDMD-mediated pyroptotic response is well-documented [36].…”

“…[34,35] Gasdermin (GSDM) pore GSDM proteins have been identi ed as a key executioner of pyroptosis. [36,37] Gasdermin D (GSDMD) forms pores in the plasma membrane, which leads to cytokine release from cells and in ammatory cell death. [38] However, GSDMD is also highly expressed in the mitochondrial membrane, where it induces the release of ROS and causes mitochondrial damage.…”

“…The GSDM proteins are predominantly expressed in the epithelium of the esophagus, stomach, colon, bladder, vagina, and prostate ( 29 , 55 ). Their presence in different tumors varies significantly, contributing to distinct roles within these tumors ( 56 , 57 ) ( Table 1 ).…”
