[Pyr 1 ]Apelin-13 Identified as the Predominant Apelin Isoform in the Human Heart

Abstract: Abstract-Apelin receptors, present on vascular smooth muscle cells, endothelium, and cardiomyocytes, are activated by the family of apelin peptides to elicit cardiovascular effects in experimental animals, but functional activity in humans has not been studied in detail. We detected low levels of apelin immunoreactivity in plasma of volunteers consistent with an autocrine/paracrine action and detected apelin immunoreactivity in the supernatant from human cultured endothelial cells. We found that [Pyr 1 ]apelin… Show more

“…A short lasting inotropic effect was also seen in isolated adult rat cardiomyocytes, where the sarcomere shortening increased for 1 -2 min only in response to apelin-16 superfusion (1-10 nM) and returned to the control before the superfusion was discontinued [17]. The different duration of the effect does not seem to be depend on the different isoforms, because (pyr)apelin-13, apelin-13 and apelin-36 show comparable potency and efficacy in inducing positive inotropic effect [8,75]. On the other hand the importance of different procedures of contractility assessment cannot be disregarded.…”

“…Apelin is reported to potentiate myocardial inotropy in isolated preparations and intact animals [74,24,8,25,17,75,41,69]. For this reason it has been suggested as a tool for the treatment of heart failure (HF) [76,70,24,67,77,78].…”
