Pycnogenol Cytotoxicity in Pancreatic INS‐1E β cells Induced by Calcium Dysregulation

Abstract: Natural standardized flavonoid extract from the bark of Pinus pinaster, Pycnogenol (Pyc), was recently found to decrease intensively the activity of sarcoplasmic reticulum Ca -ATPase of rabbit skeletal muscle (SERCA1). On the basis of this inhibitory effect in a cell-free system and similarities of SERCA1 to its other isoforms, proapoptotic properties of Pyc may be expected in cellular systems. Pycnogenol (40-100 μg/mL) induced a concentration-dependent decrease of the viability of pancreatic INS-1E β cells as… Show more

“…Rat INS-1E insulinoma pancreatic β-cells were kindly provided by Prof. Claes Wollheim, University of Geneva) and were cultured in RPMI 1640 (11 mM glucose, Sigma Aldrich) supplemented with 100 U/ml penicillin, 100 µg/ml streptomycin, 2 mM L-glutamine, 1 mM Na pyruvate, 55 µM 2-mercaptoethanol, 10 mM HEPES, 1% non-essential amino acids, and 10% fetal bovine serum, pH 7.0-7.4. The cells were grown in a humidified incubator containing 5% CO 2 at 37 °C as described previously (Viskupicova et al, 2017).…”

“…Subsequently, 100 μl of 10% sodium dodecyl sulfate in HCl (0.01 M) was added and the cells were incubated at 37 °C overnight for complete dissolution of generated formazan. The absorbance was spectrophotometrically recorded at 570 nm using the reference 690 nm (Mrvova et al, 2015;Viskupicova et al, 2017).…”

“…Pycnogenol ® (Pyc), or French maritime pine bark extract, is an extract from a plant bark known as Pinus pinaster (from class Pinopsida, order Pinales and family Pinaceae) 1 that has been studied for different purposes, such as the treatment of osteoarthritis, neuropsychiatric conditions, cardiovascular diseases, and dermato-cosmetic therapies. 2 Some of the most significant biological effects are: the protein-binding capacity, high structural mutability and function diversity. Also, the role of Pyc in the cellular antioxidant network, mediated by generegulated redox status, has been demonstrated.…”

“…Recently, the natural extract Pycnogenol 1 , obtained from the outer bark of Pinus pinaster Ait. has gained attention for its antidiabetic properties (Viskupicova et al, 2017). Pycnogenol 1 is rich in polyphenolic compounds (standardized to 70 AE 5% procyanidins) and contains catechin, taxifolin, and cinnamic/ benzoic acid derivatives (Devaraj et al, 2002) that confer a digestive enzyme inhibitory activity, especially against aglucosidase and a-amylases.…”
