Purified Human Granulocyte-Macrophage Colony-Stimulating Factor: Direct Action on Neutrophils

Abstract: Neutrophil migration inhibition factor from T lymphocytes (NIF-T) is a lymphokine that acts to localize granulocytes. Medium conditioned by the Mo human T-lymphoblast cell line was used to purify NIF-T, a glycoprotein with a molecular weight of 22,000. The NIF-T was found to potently stimulate the growth of granulocyte and macrophage colonies from human bone marrow and colony formation by the KG-1 myeloid leukemia cell line. Thus a human lymphokine (NIF-T) that modulates the activities of mature neutrophilic g… Show more

“…GM-CSF is a glycoprotein cytokine secreted by monocytes that regulate the proliferation and differentiation of myeloid cells and the activation of monocytes (32). GM-CSF has a stimulatory effect on a number of neutrophil functions (35). The IL-10 secretion was significantly increased by stimulation with GM-CSF in spleen cells compared with similarly cultured saline spleen cells (36).…”

“…94,95 The latter finding was in good accordance with the in vitro activity of GM-CSF as a potent GSD IB before and during treatment with G-CSF suggested an enhancement to the normal range in vitro and an increased inhibitor of neutrophil chemotaxis. 55,57 However, other studies showed no impairment of neutrophil migration in G-CSFinfiltration in the skin chamber assay in vivo. 13 Qualitative evaluation of migration in four patients with severe congenital and GM-CSF-treated subjects in vitro 82,96,97 and in vivo.…”

“…Proliferation of Mo-T lymphocytes (T lymphocyte cell line 20 ) was determined with a MTS colorimetric assay on a Tecan microplate reader. Mo T cells were resuspended in conditioned medium of differentiated U-937 cells to yield a final concentration of 4 · 10 5 cells/ml.…”
