Purification of multiple vitellogenins in grey mullet (Mugil cephalus)

Abstract: Three female specific serum proteins were detected immunologically in the sera of grey mullet (Mugil cephalus) which were named vitellogenin A (VgA), VgB, and VgC, based upon their distinct antigenicity against specific antisera raised against three types of mullet lipovitellins (Lvs). These Vgs were subsequently purified from the serum of estradiol-treated mullet by combining several types of chromatography columns (anion exchanger, hydroxylapatite, immunoadsorbent column, and gel filtration). Purified native… Show more

“…IgG was purified with ammonium sulfate precipitation and DE52 (Watman International Ltd. Kent, UK) ion-exchange chromatography. It is known that the grey mullet has VTG protein of three forms (A, B, C) (Amano et al, 2007). The used antibody in our experiment indicated a positive reaction to complete type of VTG-A and -B, which is the main VTG protein induced by estrogens.…”

“…To date, the complete Vtg (VtgA and/or VtgB) and incomplete Vtg (VtgC) have been successfully isolated and purified as two different proteins with distinct molecular weights and antigenicity in various fish species, including tilapia (O. aureus [85] and O. mossambicus [86]), medaka [87], Japanese goby Acanthogobius flavimanus [88] and Sakhalin taimen [89]. In addition, three distinct types of Vtg have been purified or detected in mosquitofish Gambusia affinis [90], red seabream Pagrus major [91] and grey mullet [92]. Two or three types of Vtgs have been detected as proteins with distinct antigenicity in various existing fish species [66].…”
