Purification, Characterization, and Submitochondrial Localization of the 32-Kilodalton NADH Dehydrogenase from Maize

Abstract: Plant mitochondria have the unique ability to directly oxidize exogenous NAD(P)H. We recently separated two NAD(P)H dehy-drogenase activities from maize (Zea mays 1.) mitochondria using anion-exchange (Mono Q) chromatography. The first peak of activity oxidized only NADH, whereas the second oxidized both NADH and NADPH. In this paper we describe the purification of the first peak of activity to a 32-kD protein. Polyclonal antibodies to the 32-kD protein were used to show that it was present in mitochondria fro… Show more

“…Recently, it has been suggested that in beetroot mitochondria external rotenone‐insensitive NAD(P)H oxidation is catalysed by either a 58 kDa and/or 32 kDa protein whilst matrix rotenone‐insensitive NADH oxidation occurs via a 43 kDa dimer [8–10]. Sonication studies have shown that both the matrix and external enzymes are peripherally bound to the inner‐membrane of plant mitochondria.…”

“…Antibodies were prepared against the 58-kD protein from red beet roots and used in immunoprecipitation experiments under conditions as described by Knudten et al (1994). The antibodies were incubated with maize Mono Q fractions containing the 58-kD NAD(P)H DH activity and found to immunoprecipitate 44% of the activity.…”

“…Furthermore, it is not known whether all of these enzymes exist in Arabidopsis. A number of attempts at purification of these proteins have provided little consensus as to the size and number of polypeptides involved (Klein and Burke, 1984;Luethy et al, 1991;Rasmusson et al, 1993;Knudten et al, 1994;Day, 1996a, 1996b).…”
