In this study, Bacillus subtilis 9407 showed a strong antibacterial activity against Acidovorax citrulli in vitro and 61.7% biocontrol efficacy on melon seedlings 4 days post inoculation under greenhouse conditions. To understand the biocontrol mechanism of B. subtilis 9407, identify the primary antibacterial compound and determine its role in controlling bacterial fruit blotch (BFB), a srfAB deletion mutant (ΔsrfAB) was constructed. The ΔsrfAB which was deficient in production of surfactin, not only showed almost no ability to inhibit growth of A. citrulli but also decreased biofilm formation and reduced swarming motility. Colonization assay demonstrated that B. subtilis 9407 could conlonize on melon roots and leaves in a large population, while ΔsrfAB showed a four- to ten-fold reduction in colonization of melon roots and leaves. Furthermore, a biocontrol assay showed that ΔsrfAB lost the biocontrol efficacy. In summary, our results indicated that surfactin, which consists of C13- to C16-surfactin A was the primary antibacterial compound of B. subtilis 9407, and it played a major role in biofilm formation, swarming motility, colonization and suppressing BFB. We propose that the biocontrol activity of B. subtilis 9407 is the results of the coordinated action of surfactin-mediated antibacterial activity and colonization. This study reveals for the first time that the use of a B. subtilis strain as a potential biological control agent could efficiently control BFB by producing surfactin.

Background Verticillium dahliae is one of the most destructive pathogens in many cotton-growing countries around the globe. There is still the lack of high-resistant upland cotton cultivars against this pathogen, and the usage of fungicides is threatening the environment. Biocontrol is an environment-friendly and a green technique in plant protection. In this regard, the present work was aimed to investigate antagonistic bacterial strains and to introduce them as new bioinoculants against V. dahliae in cotton. Main body of the abstract Numerous bacteria were isolated from cotton roots, and their antagonism was examined against V. dahliae. Strain SZAD2 which was identified as Bacillus velezensis on the bases of 16S rRNA gene analysis had significantly high biocontrol potential both in plate assay and under greenhouse conditions. The biocontrol efficacy in seed treatment was 60.31%, while in the soil drench method, it was 79.19%. Transmission electron microscopy (TEM) analysis revealed that the strain systemically colonized the roots and lived within the intercellular space. Hydrogen peroxide staining exposed that the strain increased plant basal defense response by the accumulation of hydrogen peroxide (H2O2) in both roots and leaves. Furthermore, the bacterization of cotton roots triggered induced systemic resistance (ISR) in the roots and caused the activation of the antioxidant enzymes such as phenylalanine ammonia-lyase (PAL), polyphenol oxidase (PPO), peroxidase (POD), and phenol contents. Moreover, the antifungal extract produced by the strain contained the activity of both cellulase and chitinase, which were able to suppress fungal mycelia both in the presence and absence of cotton. Short conclusions Strain SZAD2 systemically colonized the plant, showed multiple modes of action against the pathogen which played a vital role in V. dahliae control, and is recommended for commercial exploration as a safe alternative bioinoculant instead of hazardous chemical fungicides.

Metabolit sekunder asal mikrob merupakan sumber senyawa penting untuk pengembangan senyawa anticendawan pada strategi pengendalian penyakit. Penelitian ini bertujuan menguji kemampuan produksi metabolit sekunder bakteri antagonis rizosfer Bacillus cereus 11UJ terhadap cendawan patogen padi Rhizoctonia solani dan Pyricularia oryzae. Efek antagonis ekstrak kasar B. cereus 11UJ diuji dengan metode cakram kertas saring steril pada medium PDA. Ekstrak bakteri dengan pelarut etil asetat menunjukkan aktivitas anticendawan yang lebih baik terhadap P. oryzae dibandingkan dengan R. solani. Efek penghambatan filtrat membuktikan potensi B. cereus 11UJ untuk aktivitas anticendawan. Analisis pirolisis gas kromatografi spektrometri massa menunjukkan bahwa B. cereus 11UJ menghasilkan 3 senyawa utama, yaitu 9,19-cyclolanostan-3-ol, acetate, (3.beta.)-(CAS) cycloartanyl acetate (13.14%); 4-(2',2'-dimethyl-6'-methyliden-1'-cyclohexyliden)-3-methyl-2-butanone (9.72%); dan stigmast-5-en-3-ol, oleat (9.09%) yang diduga berpotensi menekan pertumbuhan patogen.
