Purification and Separation of Follicle Stimulating Hormone (Fsh) and Luteinizing Hormone (Lh) From Human Postmenopausal Gonadotrophin (Hmg)

Abstract: A new approach to the preparation of biologically pure FSH from HMG is described. The method entails selective binding of the LH to antibodies against HCG. The bound LH was separated from FSH by DEAE-C chromatography. An FSH preparation was obtained with a potency of 329.7 IU-FSH/mg and undetectable LH activity at the highest dose tested. This preparation, however, was not homogeneous in the immuno-electrophoretic analysis. The nature of the LH activity »unseparable« from FSH by physico-chemica… Show more

“…The results presented in this report indicate that human urinary FSH pre¬ paration promoted follicular growth to stages similar to those induced by mixtures of FSH and LH. The data presented in our previous report (Donini et al 1966), together with the histological evidence of this report, supports the hypothesis that FSH, in the absence of LH, is incapable of initiating steroid production which stimulates uterine growth.…”

“…In a recent report (Donini et al 1966), we described an immunochemical pro¬ cedure for the preparation of a biologically pure follicle stimulating hormone (FSH). The method entailed selective binding of luteinizing hormone (LH) to antibodies against human chorionic gonadotrophin (HCG) and subsequent separation of the antibody-bound LH from the FSH.…”

“…Previously derived equivalency factors (Reichert & Parlow, 1964;Donini, Puzzuoli, D'Alessio, Lunenfeld, Eshkol & Parlow, 1966) were used to compare the potencies of the three preparations. In the Steelman-Pohley assay the potency of the human FSH preparation was approximately 29 times and the porcine preparation was ten times that of ovine FSH.…”
