Purification and Physicochemical Characteristics of African Swine Fever Virus

Abstract: SUMMARYMethods for the purification of African swine fever virus (ASFV) and its dissection into two fractions are described. The difficulties which have been encountered previously in attempts to purify the virus, namely contamination with large amounts of cellular constituents and aggregation of the virus particles, have been overcome by treatment with Tween 8o and by the use of I i-NaC1 in the sucrose gradients. Five major polypeptides, tool. wt. Ioax I25 (VP1), 76 (VP2), 50 (VP3), 44 (VP4) and 39 (VPs) were… Show more

“…The number of polypeptides does not agree with that given by others (3,22), probably because they worked with extracellular virus, which cannot contain some proteins such as VP 15, a major component of intracellular virus. The low concentration of most of the minor polypeptides might be hidden in the high background of densitometry (3).…”

“…Previous studies of ASFV proteins and antigen-antibody reactions have used virus or viral proteins produced in non-porcine cells (19) or porcine cells not usually considered to be the host of viral replication in vivo (2). Recent studies of ASFV DNA revealed extensive changes in Sma I and Eeo 1%I restriction patterns associated with adaptation of the virus to growth in Veto cells (20).…”

“…Available information from polyacrylamide gel electrophoresis suggests that the number of polypeptides present in virus particles of ICDVs is quite different and not correspondingly related to virus size. African swine fever virus apparently contains only five structural proteins (Black and Brown, 1976). Frog virus 3 contains about 20 structural polypeptides Goorha and Granoff, 1974a;Willis et al, 1977), the most prevalent having a molecular weight of 55,000 and accounting for nearly 26% of the total proteins.…”
